Elemental analysis of an ionic distribution in rat basophilic leukemia cells (RBL-2H3) by in-air micro-PIXE

Rat basophilic leukemia cells (RBL-2H3) were analyzed by an in-air micro-PIXE (Particle induced X-ray emission) technique. RBL-2H3 cells, experimental model for mucosal mast cells, were used to investigate the mechanism of the aggregation of the IgE receptors, which initiates the increase of intracellular calcium concentration, and finally causes histamine release. We investigated and characterized the antigen-mediated distribution change in RBL-2H3 cells from the elemental distribution analyses by the in-air micro-PIXE technique. Sodium calcium and magnesium ions were accumulate into cells and potassium ions were run out cells after the stimulation. Interestingly, some sulfur was also run out of the cells after the stimulation. Histamine coexists with heparin in granules of the cells. In the process of histamine release, heparin might be released with granules.
