パーコレーション転移に伴う蛋白質水和水の水素結合ダイナミクスの変化

蛋白質の水和研究のモデル蛋白質である、スタフィロコッカルヌクレアーゼを用いて、0.1$$sim$$0.6(g水/g蛋白質)のいくつかの水分量で、分子シミュレーションと中性子非弾性散乱を行い、水和水の水素結合ダイナミクスや拡散を調べた。その結果、水和水の個々の水素結合ダイナミクスは、水分量に関係なく局所的な水素結合の形成パターンによって決まることが分かり、また蛋白質表面での水和水のパーコレーション転移によって、水和水全体のダイナミクスが劇的に変化することが分かった。このパーコレーション転移に伴う水和水ダイナミクスの変化は、中性子非弾性散乱で求めた拡散定数の水分量変化からも確認した。また、水和水の間の水素結合ダイナミクスは、蛋白質と水和水の間の水素結合ダイナミクスと相関があり、両者は動的にカップルしていることが分かった。水和水の水素結合ネットワークのパーコレーション転移は、水和水の物理化学的特性を決定付ける重要な要因であるとともに、生理的機能発現に重要とされる蛋白質の熱揺らぎを誘導していると考えられる。

Water network pattern and dynamics are determined by hydrogen bonding. The physical properties of the hydration water is different from that of bulk water. The hydration affects the protein biological function. Here, we show the importance of hydration for protein dynamics and the percolation of hydration water by inelastic neutron scattering and molecular dynamics simulation. The dynamical transition of protein is highly correlated with the percolation transition of the hydration water, and then the percolation induces the protein dynamical transition. The dynamical coupling between protein and hydration water is essential for protein dynamics and its biological function.
