Dose-volume histogram analysis of slit-beam irradiation; Biological implications in mouse ex vivo testes

空間的分割照射であるスリットビーム照射(SI)は、バイスタンダー効果などの非標的効果(NTE)を介して正常組織を温存できる可能性がある。しかしながら、先行研究で報告される組織温存効果(TSE)が、生物学的機構によるものか、物理的な線量体積特性に起因するものなのかは明らかではない。本研究では、既報のexvivoマウス精巣実験を対象に、放射線挙動解析コード(PHITS)を用いて、スリット幅12.5、50、および200
mに対する線量分布を算出し、生物効果を評価した。その結果、等価均一線量(EUD)は照射細胞、非照射細胞、全細胞集団に対する線量よりも、SIの評価指標として適していることが示された。これらの結果は、従来SIにおいて観察されたTSEが、DVHの差異によって説明され得る可能性を示唆している。
Slit-beam irradiation (SI), a form of spatially fractionated irradiation, has been suggested to spare normal tissue through non-target effects (NTE), such as the bystander effect. However, it remains unclear whether the tissue-sparing effects (TSE) reported in previous studies arise from biological mechanisms or from physical dose volume characteristics. In this study, we analyzed a previously reported ex vivo mouse testis experiment using the PHITS code to calculate dose distributions for slit widths of 12.5, 50, and 200
m and to evaluate the corresponding biological effects. The results indicate that the Equivalent Uniform Dose (EUD) is a more appropriate metric for characterizing SI than the doses to irradiated cells, non-irradiated cells, or the entire cell population. These findings suggest that the TSE observed in previous SI studies may be explained by differences in dose-volume histograms (DVHs).
