Labeling of bifunctional chelating agent, MAG3, with carrier-free $$^{188}$$Re

The radioisotopes of rhenium ($$^{186}$$Re and $$^{188}$$Re) are attractive radionuclides for radiotherapy because of their energetic beta particles and gamma rays suitable for imaging. Mercaptoacetyltriglycine, MAG3 (N$$_{3}$$S ligand), is a useful bifunctional ligand in labeling monoclonal antibodies with metallic radionuclides. In this study, the labeling of MAG3 with carrier-free $$^{188}$$Re from a $$^{188}$$W/$$^{188}$$Re generator was investigated in detail. The $$^{188}$$Re-MAG3 complex was synthesized by the direct labeling method and by the indirect labeling method using a transfer ligand (citric acid or gluconic acid). The dependence of the labeling yield upon the reaction conditions such as the concentrations of tin(II) chloride dihydrate as a reducing agent, S-benzoyl MAG3 and the transfer ligand, pH, temperature, reaction time and the addition of a carrier was examined. The labeling yield of $$^{188}$$Re-MAG3 synthesized by the all method was over 90% under the optimum conditions.

Applicability of Mo adsorbent PZC(Poly Zirconium Compound) for $$^{188}$$W/$$^{188}$$Re generator was investigated. Long term stability of adsorption of $$^{188}$$W to the PZC column, elution of $$^{188}$$Re from PZC column, desorption of $$^{188W}$$ from PZC column, and labeling of Hydroxyethylidene Diphosphonic Acid(HEDP) and Mercaptoacetyltriglycine(MAG3) with $$^{188}$$Re eluted from PZC column were tested. The PZC generator gave reproducible $$^{188}$$Re elution yields with low $$^{188}$$W parent breakthrough for a long period of time(about 5 months), that is the $$^{188}$$W/$$^{188}$$Re generator using PZC has a potential for practical use.

The $$beta^{-}$$ emitter $$^{177}$$Lu is a promising therapeutic radioisotope for the treatment of cancer. It has a half-life of 6.73 days and maximum $$beta^{-}$$ energy of 498 keV, resulting in a short range of radiation in tissue. The decay is accompanied by the emission of low energy $$gamma$$-radiation with $$E_{gamma}$$ = 208 keV (11.0%) and 113 keV (6.4%) suitable for simultaneous imaging. Lutetium-177 can be usually produced at nuclear reactors with high yield and high specific radioactivity by the $$^{176}$$Lu(n,$$gamma$$)$$^{177}$$Lu reaction. However, radioisotopes with higher specific radioactivity are required in the field of radioimmunotherapy using labeled monoclonal antibodies. Thus, an alternative production route, namely the $$^{176}$$Yb(n,$$gamma$$)$$^{177}$$Yb $$rightarrow$$ $$^{177}$$Lu process was studied to produce no-carrier-added (nca) $$^{177}$$Lu in this work. The radiochemical separation of the nca $$^{177}$$Lu from the macroscopic ytterbium target was investigated by means of reversed-phase ion-pair HPLC. The nca $$^{177}$$Lu was obtained in radiochemical pure form with a separation yield of 80%.

Experimental cross sections for the reactions $$^{186}$$W(d,2n)$$^{186}$$Re, $$^{nat.}$$W(d,xn)$$^{181-184}$$Re and $$^{186}$$W(d,p)$$^{187}$$W are measured up to 34 MeV using the stacked foil method. The values obtained are compared with cross sections calculated using the ALICE code. The experimental and theoretical excitation functions are in reasonable agreement except for the $$^{186}$$W(d,p)$$^{187}$$W reaction.

Adsorption of $$^{188}$$Re complexes with aminomethylenephosphonate on hydroxyapatite

The radioactive rhenium complexes with phosphonate-containing ligands are good bone seeking compounds, and have been developed for the palliative treatment of metastatic bone pain. In this paper, we describe the adsorption properties of newly developed $$^{188}$$Re complexes with aminomethylenephosphonate ($$^{188}$$Re-EDTMP, EDBMP and NTMP) on hydroxyapatite, which is the major constituent of the hard tissues of bone. The influence of the shaking time, pH and the ionic strength on the adsorption was investigated. The adsorption of $$^{188}$$Re complexes was strongly affected by pH and ionic strength in a solution. The adsorption coefficient increased with decreasing pH and ionic strength. These observations suggest that one of the causes of the adsorption of $$^{188}$$Re-amp is considered the electrostatic forces between the negative $$^188$$Re-amp complex and the positive HAP surface. The adsorption affinity of the $$^{188}$$Re-amp complex for HAP increased in the order $$^{188}$$Re-EDBMP$$<^{188}$$Re-NTMP$$<^{188}$$Re-EDTMP.
