High-intensity laser and thin-foil interactions produce high-energy particles, hard X-ray, high-order harmonics, and terahertz (THz) radiation. A proton beam driven by a high-intensity laser has received attention as a compact ion source for medical applications. In this study we have tested simultaneous generation of protons and THz radiation from a thin-foil target. We use a Ti:sapphire laser system (J-KAREN) at JAEA. A laser beam is focused by an off-axis parabolic mirror at the thin-foil target. We observed the high-energy proton in the rear side of the target and THz radiation in the reflected direction. Next, high energy protons are observed by reducing the size of preformed plasma.

Recent progress in the energy recovery linac project in Japan

Future synchrotron light source using a 5-GeV energy recovery linac (ERL) is under proposal by our Japanese collaboration team, and we are conducting R&D efforts for that. We are developing high-brightness DC photocathode guns, two types of cryomodules for both injector and main superconducting (SC) linacs, and 1.3 GHz high CW-power RF sources. We are also constructing the Compact ERL (cERL) for demonstrating the recirculation of low-emittance, high-current beams using above-mentioned critical technologies.

To further understand the catalytic mechanism and inhibitor recognition of HIV-1 protease, we need to determine the locations of key hydrogen atoms in the catalytic aspartates Asp25 and Asp125. The structure of HIV-1 protease in complex with transition-state analog KNI-272 was determined by combined neutron crystallography at 1.9 ${AA}$ resolution and X-ray crystallography at 1.4 ${AA}$ resolution. The resulting structural data shows that the catalytic residue Asp25 is protonated and that Asp125 is deprotonated. The proton on Asp25 makes a hydrogen bond with the carbonyl group of the allophenylnorstatine group in KNI-272. The deprotonated Asp125 bonds to the hydroxyl proton of Apns. The results provide direct experimental evidence for proposed aspects of the catalytic mechanism of HIV-1 protease; and can therefore contribute substantially to the development of specific inhibitors for therapeutic application.

High-intensity laser and thin-foil interactions produce high energy ions, electrons, hard X-ray, and terahertz radiation. The simultaneous generation of the protons and terahertz radiation will provide us unique applications. In this study we use a Ti:sapphire laser system (JLITE-X) at JAEA for simultaneous generation of the protons and terahertz radiation. The laser beam is focused by an off-axis parabolic mirror at the Ti target surface. The estimated peak intensity is $$sim$$2$$times$$10$$^{17}$$W/cm$$^{2}$$. The simultaneous generation of the protons and terahertz radiation are observed by reducing the ASE duration.

At present, using ultra-short high intensity lasers at APRC, JAEA Kansai photon research institute, we are developing laser driven multiple quantum beams such as protons, X-rays, electrons and THz waves. These beams are perfectly synchronized with each other. The pulse duration of each beam is lass than a pico-second. They have sharp directionality with high brightness. If we properly combined these, we have new pump-probe techniques for various applications.

We have determined a crystal structure of HIV-1 protease by neutron crystallography. The development of HIV-1 protease inhibitors is regarded as a major success of structure-based drug design and contributes to establish highly active anti-retroviral therapy for AIDS. To further understand the catalytic mechanism of HIV-1 protease and interaction between HIV-1 protease and its inhibitor, we have determined the crystal structure of HIV-1 protease in complex with a inhibitor, KNI-272 to 2.3 ${AA}$ resolution by neutron crystallography. Our results indicates that the carbonyl group of allophenylnorstatine (Apns) in KNI-272 forms a significant hydrogen bond with protonated Asp 25, and the hydrogen atom from the hydroxyl group of Apns forms a remarkable hydrogen bond with the deprotonated Asp125. These results show direct evidence that Asp25 provides a proton to carbonyl group of substrate and Asp125 contributes to activate the attacking water molecule as a nucleophile.

High-intensity laser and thin-foil interactions produce high energy ions, electrons, X-ray, high-order harmonics, and THz radiation. High-energy protons driven by the high-intensity laser is paid attention as a compact ion source for medical application. The simultaneous generation of the protons and THz or harmonics will provide us high-density plasma diagnostic or unique pump-probe techniques. We use a Ti:sapphire laser system (JLITE-X) in JAEA for THz radiation. The laser beam is focused by an off-axis parabolic mirror at the 5 $$mu$$m thick Ti target. We observe simultaneously both the high-energy proton and THz radiation by changing the duration of ASE preceding the main pulse. We use a Ti:sapphire laser system in CRIEPI for harmonic generation. The laser beam is focused by an off-axis parabolic mirror at the 7.5 $$mu$$m thick polyimide target surface. The high-energy protons and UV harmonics are observed at the target is moved away from the best focus.

HIV-1 protease is a dimeric aspartic protease that cleaves the nascent polyproteins of HIV-1 and plays an essential role in viral replication. To further understand the catalytic mechanism of HIV-1 protease, we have determined the crystal structure of HIV-1 protease in complex with a transition state mimetic tripeptide inhibitor, KNI-272 to 1.9 ${AA}$; resolution by neutron crystallography in combination with 1.4 ${AA}$; resolution X-ray diffraction data. Our results indicates that the carbonyl group of allophenylnorstatine (Apns) in KNI-272 forms a significant hydrogen bond with protonated Asp 25, and the hydrogen atom from the hydroxyl group of Apns forms a remarkable hydrogen bond with the deprotonated Asp125.
