本研究では、土壌環境微生物を対象として腐食ポテンシャルおよびそのメカニズムについて知見を得るために、一般的な湖沼底泥を植菌源として炭素鋼とともに培養を行った。その結果、いずれの培養条件でも炭素鋼を腐食、減損する活性が見られ、特に有機物添加・汽水培地において硫酸還元活性、メタン生成活性とともに高い腐食活性が得られた。土壌埋設設備への微生物の腐食影響を適切に評価するためには、硫酸還元菌, メタン生成菌などの相互作用を踏まえた評価が必要と考えられる。

地下微生物生態研究の進展により、地下数km以深における微生物の存在が明らかとなり、地層処分における微生物影響評価は重要な課題となっている。本研究では、地層処分研究開発に関する「第2次取りまとめ」以降に刊行された、国内外の主要な学術論文や報告書等の文献約200件を精査し、高レベル放射性廃棄物だけでなく、TRU併置や使用済み核燃料の直接処分も含め、地層処分システム性能に影響を及ぼす微生物要因に関する最新の知見を取りまとめた。

本発表では、地層処分システムにおいて人工バリアの閉じ込め性能に及ぼす微生物影響について、これまでの国内外での検討事例を紹介する。また、日本で検討されている緩衝材であるクニゲルV1およびケイ砂の混合物を対象とした微生物の金属腐食影響について、硫酸還元菌だけでなくメタン菌や酢酸生成菌などの腐食菌存在下において実施した試験結果について報告する。圧縮ベントナイトの乾燥密度1.0および1.6g/cm、温度条件30Cおよび50Cにて、多様な種を含む腐食微生物群集の存在下で炭素鋼を用いた腐食試験を実施した結果、一年間の試験期間内では、50Cおよび乾燥密度1.6g/cmの条件下では腐食が抑制されていたが、30Cおよび1.0g/cmの条件下では6ヶ月から1年の間に腐食が急速に進むことが確認された。緩衝材の圧縮密度が低下した場合には、微生物腐食が短期間で顕著に進む可能性が示唆された。

Radioactive waste from nuclear power generation would be overpacked in metallic material, such as carbon steel. The lifetime of metallic containment is strongly related to the corrosion rate of carbon steel in compacted buffer material. Corrosion of carbon steel buried in compacted buffer material with different dry densities in test cells was investigated in lab experiments using corrosive microbial consortium. Compacted buffer materials were incubated in nutrient medium with or without inoculation by an enriched microbial community for one year at 30C and 50C. Corrosion rates were suppressed (13.6 - 18.6 mg/year) in samples with compacted densities of 1.3 and 1.6 g/cm at 50C. In contrast, a high corrosion rate was observed in the inoculated 1.0 g/cm compacted bentonite buffer materials at 30C. Although the corrosion rate was low until the sixth month (4.6 mg/year), it significantly increased to 74.9 mg/year after one year. It is demonstrated that a sufficiently high dry density is one of the important key factor to suppress microbial activity in buffer material surrounding metal containers, because of the physical characteristics such as small pores, low water activity, less nutrient supply caused by low hydraulic conductivity.

Microbial activity in bentonite buffer materials in deep geological repository is of concern for microbiologically influenced corrosion which could affect the longevity of metallic containers. Data has been collected regarding microbial corrosion of carbon steel or cupper using compacted bentonite. However, there is not enough data collected under Japanese geological disposal concept, using Kunigel V1 and silicate sand. To evaluate microbial effects on corrosion of metallic containers and activity in bentonite buffer materials with Kunigel V1, we have conducted laboratory-scale and in-situ experiments. From the result of the experiments, it is demonstrated that a sufficiently high dry density is one of the important key factors to suppress microbial activity in buffer materials surrounding metal containers, because of the physical characteristics such as small pores, low water activity, less nutrient supply caused by low hydraulic conductivity. However, microbial cells were heterogeneously detected in the bentonite buffer materials of in-situ experiment. This seems to result from nonuniformity in dry density, water content, and swelling pressure etc., in the engineering scale experiment. These results also demonstrate that the heterogeneous repository conditions would be possibly occurred.

Microbially influenced corrosion of metallic container is one of concerns for nuclear waste disposal. Corrosion of carbon steel buried in compacted bentonite buffer materials was investigated using anaerobic corrosive microbial consortium. The buffer material was composed of 70% bentonite (Kunigel V1) and 30% silicate sand with different dry densities in test cells. Compacted buffer materials were incubated in nutrient medium with or without inoculation for one year at 30C and 50C. Corrosion rates were suppressed (9.4-12.9 mg/cm/year) with compacted densities of 1.3 and 1.6 Mg/m at 50C. In contrast, a high corrosion rate (52.0 mg/cm/year) was observed in the inoculated 1.0 Mg/m compacted buffer materials at 30C, and microbial analysis showed the highest microbial abundance and the high relative ratio of sulfate-reducing bacteria, such as sp.. These results are demonstrated that a sufficiently high dry density is one of the important key factors to suppress microbial activity in buffer material surrounding metallic containers, because of the physical characteristics such as small pores, low water activity, less nutrient supply caused by low hydraulic conductivity.

深部地下に放射性廃棄物を埋設する地層処分事業では、放射性廃棄物の閉じ込めを確実とするため放射性廃棄物を地下に埋設することが検討されている。深部地下においても多様な微生物が存在するため、この人工バリアの閉じ込め性能を評価する上で、微生物による金属製処分容器の腐食影響を考慮する必要性がある。そこで、本研究では異なる密度条件で圧縮したベントナイト内に設置した炭素鋼の微生物による腐食影響を解析した。その結果、緩衝材中の微生物活性には圧縮密度と温度が重要な条件であるとともに、硫酸還元菌がベントナイト内での炭素鋼の腐食に寄与していることが示唆された。また、嫌気性腐食微生物群集に含まれる硫酸還元菌による炭素鋼腐食過程を解析するために、電気培養槽において炭素鋼表面での腐食電位、電流変化を経時的に計測したところ、電位は全ての試料で培養開始後急激に低下し、無菌区はほぼ安定した値を示したが、植菌区では培養1週間後に増加に転じた。また、炭素鋼表面ではDesulfovibrioの増加が見られたことから、これらの電位の上昇はDesulfovibrioによるcathode電流の利用反応促進によるものと推定された。処分環境においては、処分後100年程度までは約70C以上の高温環境下であることが予測されており、微生物増殖に適した環境ではないと考えられるが、閉じ込め性能における微生物影響の不確実性を排除するために、今後、緩衝材中の硫酸還元菌を含めた微生物活性を抑制する要因(膨潤圧・水分活性・空隙サイズ・温度等)の詳細な検討が必要である。
