The SNARE protein SYP71 expressed in vascular tissues is involved in symbiotic nitrogen fixation in ${it Lotus japonicus}$ nodules

Rhizobial symbiotic nitrogen fixation in root nodules is regulated by the host legume genes. Fix- mutants that exhibit lower or no nitrogen-fixation activity are useful to identify host plant genes required for symbiotic nitrogen fixation. Here, we show a ${it Lotus japonicus}$ novel Fix- mutant defective of a SNARE protein. The mutant formed nodules that displayed lower nitrogen fixation activity, and the growth of the host plant was retarded. Exogenous combined nitrogen almost recovered the growth of the mutant. Numbers of nodules formed on the mutant were similar to those on the wild-type plant. However, the mutant nodules were smaller and showed early senescence. The causal gene was identified by map-based cloning, and the predicted protein was appeared to be homologous to one of SNARE proteins found in Arabidopsis thaliana. The responsible gene was expressed ubiquitously in shoot, roots and nodules. In roots and nodules, the transcripts were detected in vascular bundles. These results indicated that a SNARE protein expressed in vascular tissues is required for nitrogen fixation activity of rhizobia in nodules.

マメ科植物は共生根粒の発達及び数を正確に制御している。ミヤコグサ(${it Lotus japonicus}$)での根粒形成の制御について、地上部に存在する${it HAR1}$タンパクが必要であることから地上部と根の間での長距離の情報伝達経路の存在が示唆されている。この情報伝達経路の分子機構に対する理解を深めるため、われわれは(${it too much love}$ (${it tml}$))と命名した超根粒着生変異体を単離し解析した。野生型との接木試験によって${it tml}$変異体での過剰な根粒形成は根部の遺伝子型に依存することが示された。${it har1}$変異体の地上部と${it tml}$変異体の根部の接木では根粒の着生に影響は見られなかった。これらの結果は地上部の因子である${it HAR1}$と根の因子である${it TML}$が同じ情報伝達経路に関与することを示している。${it TML}$${it HAR1}$の下流で機能しており、${it TML}$が地上部から根に輸送される何らかのシグナル因子の受容体として機能している可能性がある。

300Gyのヘリウムイオンビーム照射によって高根粒形成能力を持つ新規のミヤコグザの突然変異体Klavier(klv)が得られた。ミヤコグザでは、根粒形成が減少する突然変異体は多く得られるが、本突然変異体のように、根粒形成が増進されるのは非常に稀である。klv突然変異体は、根の周りに小さな根粒を非常に多数形成するばかりでなく、葉脈の形成に異常が観察され、また花期が遅れる特徴を持ち、さらに矮化変異を伴っていた。しかしながら、染色体マッピングにより、klv遺伝子の同定を試みたところ、第一染色体の長腕の0.29cM以内の位置にマップできることがわかり、単一で劣性の遺伝子であることがわかった。このことから、根粒形成と維管束や花器形成には何らかの関連があることが示唆された。

植物はDNA変異原の曝露により生じるDNA損傷の影響を回避するために、さまざまな機構を備えている。DNA損傷乗り越え複製(TLS)はその一つである。近年、誤りがちなDNA損傷乗り越え複製(error-prone TLS)にかかわる、DNAポリメラーゼ$$zeta$$のサブユニット${it REV3}$のシロイヌナズナホモログ(${it AtREV3}$)の遺伝子破壊系統rev3が単離,解析され、植物でもerror-prone TLSの存在が示唆された。本研究では、error-prone TLSで機能していると予想される、シロイヌナズナ${it AtREV1}$, ${it AtREV7}$遺伝子の遺伝子破壊系統(${it rev1}$, ${it rev7}$)の単離,解析を行い、植物のerror-prone TLSについてより詳細な解析を試みた。${it rev1}$は、UV-B, DNA架橋剤に対して感受性を示し、${it rev3}$株のDNA変異原に対する感受性と傾向が似ていた。一方で${it rev7}$は長期間のUV-B, DNA架橋剤に対して感受性を示した。この結果は${it AtREV1}$, ${it AtREV7}$が程度の違いはあるがDNA変異原によって誘導された各種DNA損傷の回避に関与していることを強く示唆しており、植物にも他の生物と同様のerror-prone TLSが存在すると考えられた。
