Ion beam irradiation has different influences on superoxide dismutase activity in cultured human retinal vascular endothelial cells between $$^{12}$$C and $$^{4}$$He

It is known that superoxide dismutases (SOD) are a class of enzymes that catalyze the dismutation of superoxide into oxygen and hydrogen peroxide. Ionizing radiation is known to induce oxidative stress through generation of reactive oxygen species resulting in imbalance of the pro-oxidant and antioxidant in the cells, which is suggested to culminate in cell death. Therefore, we measured the activity of SOD in human RE cells exposed to the He- and C-ion beam. The cells collected at 0, 4, 8, and 24 hr after irradiation were extracted by adding the SOD assay buffer to the pellets, and lysed by several cycles of freezing and thawing. The activity of SOD was measured using a modification of the luminol assay. SOD activity decreased according to duration time after irradiation of He-ion, however, in case of C-ion, it increased at 24 hr after irradiation. The result suggested that that there were the differences of the effects by irradiation on SOD activity between He- and C-ion.

We examined the influence of L-dopa on the inactivation of reactive oxygen species by glutathione peroxidase (GPx), which protects in human retinal vascular endothelial (RE) cells from free radical damage. We used ion beams to induce oxidative stress and measured GPx expression by means of real-time reverse-transcriptase-polymerase chain reaction (RT-PCR) analysis. Exposure to L-dopa inhibited GPx expression in human RE cells. GPx expression in RE cells incubated with L-dopa decreased significantly after exposure to 50 MeV He and 220 MeV C, disturbing the defense mechanism against the L-dopa-induced production of reactive oxygen species. In contrast, 350 MeV Ne irradiation significantly increased GPx3 expression. The result indicated that there are the influence of ion beams on L-dopa-induced stress rather than the influence of L-dopa on ion beam-induced stress because GPx expression in L-dopa-treated human RE cells depended on the types of heavy ion used during irradiation.

We investigate how L-dopa influences glutathione peroxidase (GPX) in preventing damage to membrane phopholipids of retinal endothelial cells (RE). Human RE incubated in vitro with 250 $$mu$$M L-dopa were exposed to heavy ion beams of 350 MeV Ne, 220 MeV C, and 50 MeV He. We collected RE after 0, 4, 8, 24 hrs of irradiation, extracted total cellular RNA, and synthesized cDNA, then the expression of GPX and 18S RNA were measured by realtime RT-PCR using specifically designed primers. The exposure to L-dopa inhibited the expression of GPX in RE. The expression of GPX in RE incubated with L-dopa decreased significantly after exposure to $$^{4}$$He and $$^{12}$$C. In contrast, $$^{20}$$Ne- ion beam irradiation significantly increased the expression of GPX in RE incubated with L-dopa. The result indicated that the difference of LET of ion species resulted to the different expression GPX in RE incubated with L-dopa.
