Tumor radioresistance caused by radiation-induced changes of stem-like cell content and sub-lethal damage repair capability

Alive cancer cells after fractionated irradiations with 2 Gy X-rays per day for more than 30 days show clinically relevant radioresistant. Such radioresistance is experimentally interpreted to attributed to the increment of stem-like cell content. However, only an experimental approach cannot clarify the cell responses (DNA damage and cell death induction) of cancer stem cells, so the radioresistant mechanisms remain uncertain. In addition to the conventional cell experiments using radio-resistant cell lines established after fractionated irradiations, in this study we developed a mathematical model (so called integrated microdosimetric-kinetic (IMK) model) explicitly considering cancer stem-like cell content and DNA damage responses and investigated radioresistant mechanisms acquired after fractionated irradiations. The IMK model analysis suggested that the changes of stem-like cell fraction and DNA repair efficiency play important roles of radioresisitance acquired after irradiations. Considering these into the IMK model, we successfully reproduced the experimental survival of various cell lines and various irradiation conditions. This work would contribute to not only the precise understanding of the radioresistant mechanisms induced after irradiation but also predicting curative effects with high precision.

A 3 MeV linac for development of accelerator components at J-PARC

We have constructed a linac for development of various accelerator components at J-PARC. The ion source is same as the J-PARC linac's, and the RFQ is a used one in the J-PARC linac. The beam energy is 3 MeV and nominal beam current is 30 mA. The accelerator has been already commissioned, and the first development program, laser-charge-exchange experiment for the transmutation experimental facility, has been started. In this paper, present status of this 3-MeV linac is presented.

Development of beam scrapers using a 3-Mev linac at J-PARC

We have used a beam scraper with the incident angle of 65deg to reduce the beam power deposition density in the MEBT between a 324 MHz RFQ and a 50-MeV DTL of the J-PARC linac. The 65$$^{circ}$$ scraper was irradiated by the H$$^{-}$$ beam up to particle number of 1.47E22. We observed a lot of surface projections with several hundred micrometers high in the beam irradiation damage on the scraper by using the laser microscope. In order to study the limits of scrapers, we constructed a new 3 MeV linac at J-PARC. We will conduct the scraper irradiation test at the end of this year.

On the radioactive waste disposal, the long-term mechanical stability of shafts and galleries excavated in rock is required. Therefore, it is very important to understand the time-dependent behavior of rock mass for evaluating long-term mechanical stability. The purpose of this study is determining the mechanisms of time-dependent behavior of rock mass by precise testing, observation and measurement in order to develop methods for evaluating long-term mechanical stability of a rock mass. This report describes the results of the activities in fiscal year 2013. In Chapter 1, we described the overview and background of this study. In Chapter 2, the results of a long-term creep test on Tage tuff, started in fiscal year 1997 are described. In Chapter 3, the result of organization and analysis for time-dependent behavior of crystalline rock was described. In Chapter 4, for the drafting of in-situ test plan, examination of the numerical analysis technique of rock mass was carried out.

It is very important to understand the time-dependent behavior of rock for evaluating long-term mechanical stability. The purpose of this study is determining the mechanisms of time-dependent behavior of rock by the precise test, observation and measurement, to develop methods for evaluating long-term mechanical stability. This report describes the results of the activities in fiscal year 2012. In Chapter 1, we provide an overview and the background to this study. In Chapter 2, the results of a long-term creep test on Tage tuff, started in fiscal year 1997 are described. In Chapter 3, the experimental results concerning the loading-rate dependency of strength were examined to understand the time-dependency of rock. In Chapter 4, the stability of tunnels, around which rock stress is larger than that around a circular tunnel, were examined to obtain useful information on the future plan for ${it in-situ}$ tests in the underground research laboratory.

With respect to high-level radioactive waste disposal, knowledge of the long-term mechanical stability of shafts and galleries excavated in rock are required, not only during construction and operation but also over a period of thousands of years after closure. Therefore, it is very important to understand the time-dependent behavior of rock for evaluating long-term mechanical stability. This report describes the results of the activities in FY2010. In Chapter 1, we provide an overview and the background to this study. In Chapter 2, the results of a long-term creep test on Tage tuff, started in FY1997 are described. In Chapter 3, the relation of loading-rate dependency of strength and stress dependency of creep life, the relationship of time dependency and probability distribution of strength or creep life and size effects are discussed to indicate more clearly the meaning of the value of "n" to express the degree of time dependency of the rock. In Chapter 4, failure criterions of a rock mass considering time dependency are discussed. In Chapter 5, the FEM analysis implemented with a generalized variable-compliance-type constitutive equation considering the change of "n" in confining pressure and strength distribution of rock mass carried out to clarify the long-term behavior of Toki granite is described. Finally, based on the numerical analysis, an in situ testing method is proposed.

Power supply system for klystron in J-PARC linac

This paper describes the present status, technical specifications, operating experience, and upgrade plane for the power supply system (PS system) for the klystrons in the J-PARC. The PS systems include both the high voltage DC power supplies (DCPSs), which drives one or four modulating-anode klystron each, and the anode-modulators, one per klystron. Currently, the energy output of the J-PARC linac is 181 MeV, which includes the energy necessary to run twenty 324 MHz klystrons. In 2012, the linac will add twenty-five 972 MHz klystrons and upgrade the energy output to 400 MeV.

Development of LLRF control software for the J-PARC 400 MeV Linac

The output energy of the J-PARC proton Linac will be upgraded from 181 to 400 MeV in the next two years by adding 972-MHz high-beta acceleration sections. The RF signals are controlled by the FPGA-based digital feedback control systems installed in a compact PCI (cPCI). Recently, the LLRF control software has also been upgraded for the J-PARC Linac, especially for the 972-MHz high-beta systems. Many functions have been added to the LLRF control software, such as (1) gradually increasing the feedback gains in the feedback loop instead of fixed ones, (2) automatic chopped-beam compensation, (3) automatically switching the beam loading compensation in accordance with the different beam operation mode, (4) input RF-frequency tuning carried out by a FPGA to match the RF cavities during the RF start-up, (5) auto-tuning of the RF cavity tuner by detecting the phase curve of the RF cavity during the field decay instead of the phase difference between the cavity input and output signals.

LLRF controller upgrade for the J-PARC 400 MeV linac

The output energy of the J-PARC proton linac will be upgraded from 181 to 400 MeV in the next two years by adding high-b acceleration sections. Structure of liquid water under high pressure up to 17 GPa

The structure of liquid water was studied along the melting curve up to 17.1 GPa and 850 K by in situ X-ray diffraction. At low pressures, the local structure changed toward a simple liquid-like structure through an increase in the coordination number of water molecules. Once densely packed structure of water molecules was achieved around 4 GPa, the volume was reduced through the decrease of the intermolecular distance on further compression. Classical molecular dynamics simulations well reproduced the experimental results although the degree of agreement depended on pressure. Limitations of the pair potential model were discussed.

The beam commissioning of the J-PARC linac started in November 2006 and 181 MeV acceleration was successfully achieved in January 2007. The linac has delivered beams to the 3 GeV Rapid Cycling Synchrotron for its commissioning, and then, the subsequent Main Ring Synchrotron and the neutron target commissioning. The linac uses a Cs-free LaB$$_{6}$$-driven ion source and 20 units of 324 MHz klystrons. As of June 2008, the operation times are about 3,000 and 6,000 hours for the ion source and the RF source, respectively. The operating experience of the linac is described.

High power operation of all the RF systems of J-PARC linac was started for the cavity conditioning in October 2006. High power RF sources have been installed in J-PARC linac building. The RF source is defined as the microwave application equipment by the Radio low, and is needed the charge minister's permission to offer the service. To pass the regulation, we sought the leakage point using a search coil and a bilog antenna, and stopped up the leak by contacting the metal connection completely. The result of measuring the microwave at the J-PARC site boundary, we confirmed the field strength which is less than permissible level.

Performance of J-PARC LINAC RF system

High power operation of all the RF systems of J-PARC linac was started for the cavity conditioning in October 2006. Twenty 324-MHz klystrons have supplied the power to the accelerating cavities successfully. The beam commissioning was started in November 2006 then the acceleration to the design energy of 181 MeV was succeeded. The injection to the 3-GeV synchrotron will be started in September 2007. The RF control systems are performing according to the expectation, and the required field stability is satisfied.
