We developed a simple and rapid technique to generate high-purity and high-yield [N]N gas and obtained real-time images of N fixation in an intact soybean plant. The purification efficiency was 81.6% after decay correction.

However, the reliable technique to reduce substantially rice Cd contamination has not been offered so far. Here, we report the ion-beam irradiated rice mutants that do not nearly accumulate Cd in the grains and the mutant gene responsible for the Cd limitation. We found three mutants in which a favorable low-Cd trait is expressed by the different mutations on the same gene (), and the transporter encoded by the mutant gene had a defective function of root Cd influx. The mutants adapted well in the Cd-polluted paddy fields by exhibiting nearly non-detectable Cd concentrations in their grains in accordance with no adverse economic traits. The DNA marker has been developed to breed new cultivars carrying . Our findings would be useful for greatly reducing the Cd level of paddy rice.

PETIS has been applied for analyses of cadmium transport from the root organ to the grains in rice plants in previous studies. In this study, we first realized direct imaging and quantitative analysis of the dynamic process of cadmium transport from hydroponic culture to shoot via root organ. When a radiotracer is applied to plant roots for imaging experiments, it has been usual that the region of application is shielded or set off from the field of view because of too high radioactivity there. We successfully obtained serial quantitative images by developing suitable vessels for observation and arranging best radioactivity of the tracer. High-Cd-accumulating lines of rice and Sedum, a hyper accumulator plant for heavy metals, were subjected to this experimental system and analyzed for their abilities of Cd uptake.
