Web page access analysis and text mining for extraction of the needs in countermeasures against the radioactive contamination

Extraction of the needs related to countermeasures against radioactive contamination was conducted by access analysis of the "Base Information Q&A Site" operated by JAEA and text mining of stenographic text data from a briefing on R&D by JAEA for local government officials. The results of the access analysis indicated that even after over 10 years had passed since the 1F accident, there were needs for data and information on the concentration of radiocesium in the environment, and agricultural, forestry, and fishery products and air dose rates used as a degree of radioactive contamination and evidence for the lifting of evacuation orders. The text mining results suggested the needs for data and information on radiocesium concentrations and air dose rates for river systems and aquatic products in the northern part of Hamadori, for forested environment and forest products in Nakadori, and for decontamination and air dose rates in forests and reservoirs in the southern part of Hamadori, as well as for monitoring data on changes in air dose rates after decontamination. The needs for information and data based on surveys and monitoring on the current situation of radioactive contamination rather than the future situation were also inferred.

We report crystal growth of human immunodeficiency virus 1 protease (HIV PR) in a complex with its inhibitor KNI-272 by six different methods. Comparative analysis indicates that top-seeded solution growth (TSSG) and TSSG combined with the floating and stirring technique (TSSG-FAST) are efficient strategies for rapidly obtaining large single crystals and effectively preventing polycrystallization of the seed crystal. Neutron diffraction analysis confirmed that the crystalobtained by TSSG is a high-quality single crystal. Furthermore, crystal shape was observed to be influenced by solution flow, suggesting that the degree of supersaturation significantly affects the crystal growth direction of HIV PR complex. This finding implies that the shape of the HIV PR complex crystal might be controlled by the solution flow rate.

A series of HIV protease inhibitor based on the allophenylnorstatine structure with various P2' moieties were synthesized. Among these analogues, we discovered that a small allyl group would maintain potent enzyme inhibitory activity compared to that of the $$o$$-methylbenzyl moiety in clinical candidate 1 (KNI-764, also known as JE-2147, AG-1776 or SM-319777). Introduction of an anilinic amino group to 2 (KNI-727) improved water-solubility and anti-HIV-1 activity. X-ray crystallographic analysis of 13k (KNI-1689) with a $$beta$$-methallyl group at P2' position revealed hydrophobic interactions with Ala28, Ile84, and Ile50' similar to that of 1. The presence of an additional methyl group on the allyl group in compound 13k significantly increased anti-HIV activity over 1, while providing a rational drug design for structural minimization and improving membrane permeability.

Safeguards guidelines for post-accident facilities and associated waste management facilities

In the consultancy meeting "Safeguards Guidelines for post-accident facilities and associated waste management facilities" held in IAEA, decommissioning activities, and the relevant safeguards activities in TRP/PCDF are reported. In general, the facility after accident with access limitation, operator decides to conduct decommissioning. Therefore, our report regarding the decommissioning plan and the relevant safeguards activities are very helpful to be considered. The nuclear material remove and waste management (storage/treatment) are essential work in the normal accessible facility as well as post-accident facility. Otherwise, in the case of post accident facility, alternative safeguards measures is required according to the access situation because normal safeguards activities might not be taken appropriately. In this report, we would present that outline of decommissioning plan in the TRP/PCDF and the relevant safeguards and nuclear material accountancy.
