The initial biomass was the highest with the flourish of aerobic H-oxidizing Hydrogenophaga spp., whereas an uncultivated lineage of the phylum Nitrospirae became predominant after three years with decreasing biomass. The common occurrence of many species of Nitrospirae and Chlorobi phyla at the geographically distinct sites and the exclusive detection of their phylogenetically related environmental sequences from deep groundwaters and terrestrial hot springs, suggest that these bacteria are indigenous and potentially adapted to the deep terrestrial subsurface.

Deep groundwater of meteoric origin was collected from underground boreholes drilled into the Cretaceous Toki granite, central Japan. A large sulfur isotopic fractionation of 20-60 permil diagnostic to microbial sulfate reduction is associated with the investigated groundwater containing sulfate below 0.2 mM. In contrast, a small carbon isotopic fractionation ( 30 permil) is not indicative of methanogenesis. Our results demonstrate that the deep biosphere in the terrestrial crust is metabolically active and playing a crucial role in the formation of reducing groundwater even under low energy fluxes.

Following FY2011, JAEA and NUMO have conducted a collaborative research work which is designed to enhance the methodology of repository design and performance assessment in preliminary investigation stage. With regard to (1) study on rock suitability in terms of hydrology, the tree diagram of methodology of groundwater travel time has been extended for crystalline rock, in addition, tree diagram for sedimentary rock newly has been organized. With regard to (2) study on scenario development, the existing approach has been improved in terms of a practical task, and applied and tested for near field focusing on the buffer. In addition, the uncertainty of some important processes and its impact on safety functions are discussed though analysis. With regard to (3) study on setting radionuclide migration parameters, the approaches for parameter setting have been developed for sorption for rocks and solubility, and applied and tested through parameter setting exercises for key radionuclides.

(thickness 20 nm) were irradiated with 120-720 keV C
ions and observed using transmission electron microscopy (TEM). The ion track produced in an amorphous material was directly observed by TEM. For quantitative analysis, the ion tracks were also observed using high-angle annular dark field scanning transmission electron microscopy (HAADF-STEM). The observed ion track consists of a low density core (radius
2.5 nm) and a high density shell (width
2.5 nm), which is very similar to the ion tracks in amorphous SiO
irradiated with high energy heavy ions observed by small angle X-ray scattering (SAXS). Although the observed ion tracks may be affected by surface effects, the present result indicates that TEM and HAADF-STEM have potential to observe directly the fine structures of ion tracks in amorphous materials.

(3) Study on setting nuclide migration parameters: the approach for parameter setting has been improved for sorption and diffusion coefficient of buffer/rock, and applied and tested for parameter setting of key radionuclides. (4) Study on ensuring quality of knowledge: framework for ensuring quality of knowledge has been studied and examined aimed at the likely disposal facility condition.
