Tetrathionate hydrolase (4THase) from the iron- and sulfur-oxidizing bacterium ${it Acidithiobacillus ferrooxidans}$ catalyses the disproportionate hydrolysis of tetrathionate to elemental sulfur, thiosulfate and sulfate. The gene encoding 4THase (${it Af-tth}$) was expressed as inclusion bodies in recombinant ${it Escherichia coli}$. Recombinant ${it Af}$-Tth was activated by refolding under acidic conditions and was then purified to homogeneity. The recombinant protein was crystallized in 20 m${it M}$ glycine buffer pH 10 containing 50 m${it M}$ sodium chloride and 33%(${it v/v}$) PEG 1000 using the hanging-drop vapour-diffusion method. The crystal was a hexagonal cylinder with dimensions of 0.2 $$times$$ 0.05 $$times$$ 0.05 mm. X-ray crystallographic analysis showed that the crystal diffracted to 2.15 ${AA}$ resolution and belongs to space group ${it P}$3$$_{1}$$ or ${it P}$3$$_{2}$$, with unit-cell parameters ${it a}$ = ${it b}$ = 92.1, ${it c}$ = 232.6 ${AA}$.

This study was undertaken to elucidate the intracellular changes of metal elements after the administration of fucoidan extracted from ${it Cladosiphon okamuranus}$. TRL1215 cells (normal rat liver cell line) were treated with 0, 0.1, or 1.0 mg/mL fucoidan. The cellular distribution of elements was analyzed using in-air micro-PIXE induced X-ray emission. In fucoidan-treated cells, the focal accumulation of Br (BrdU) was correlated spatially with phosphorous-rich region, suggesting that Br was localized within the nucleus. Al distribution provided a spatial association with Br map. These data suggest that fucoidan binding Al us postulated to be transferred into the nucleus.

Application of real time PCR for the quantitative detection of radiation-induced genomic DNA strand breaks

Yamauchi, Emiko*; Watanabe, Ritsuko; Oikawa, Miyoko*; Fujimoto, Hirofumi*; Yamada, Akinori*; Saito, Kimiaki; Murakami, Masahiro*; Hashido, Kazuo*; Tsuchida, Kozo*; Takada, Naoko*; et al.

Journal of Insect Biotechnology and Sericology, 77(1), p.17 - 24, 2008/02

The frequency of single strand breaks (SSBs) occurring on both strands of the pBR322 plasmid DNA region flanked by a pair of primers used for polymerase chain reaction (PCR) amplifications was determined after irradiation with $$^{137}$$Cs $$gamma$$-rays. We refined that real time PCR is suitable for the detection and quantitative analysis of SSBs caused by $$gamma$$-ray irradiation. The utility of this approach was also supported by the comparison of the practical experimental data with the Monte Carlo simulation. The potential application of this PCR method for the detection of genomic DNA damage was also confirmed.

A new program was started out to create a new medical scientific field, which is consisting of radiation oncology and nuclear medicine, utilizing the advanced accelerator and ion beam technology. An in-air micro-PIXE analyzer system, which is among the most important technical basis of the program, was upgraded to improve accuracy of elemental mapping for samples having thickness variation in a scope of microbeam scanning. In the program, on the other hand, in order to approach important bio-medical problems on cancer, intracellular dynamics of the trace elements according to mechanism of development of diseases were studied using this system. This paper outlines this program and shows the system upgraded, and results of preliminary studied about the problems.
