Fabrication of enzyme-degradable and size-controlled protein nanowires using single particle nano-fabrication technique

Protein nanowires exhibiting specific biological activities hold promise for interacting with living cells and controlling and predicting biological responses such as apoptosis, endocytosis and cell adhesion. Here we report the result of the interaction of a single high-energy charged particle with protein molecules. Degradation of the human serum albumin nanowires was examined using trypsin. The biotinylated human serum albumin nanowires bound avidin, demonstrating the high affinity of the nanowires. Human serum albumin-avidin hybrid nanowires were also fabricated from a solid state mixture and exhibited good mechanical strength. The biotinylated human serum albumin nanowires can be transformed into nanowires exhibiting a biological function such as avidin-biotinyl interactions and peroxidase activity. The present technique is a versatile platform for functionalizing the surface of any protein molecule with an extremely large surface area.

An Ion micro-Beam-Induced Luminescence (micro-IBIL) system was newly developed for the evaluation of the accuracy of proton beam writing on solid diamond surface. Fabrication of buried microstructures in bulk diamond (3.0 mm $$times$$ 3.0 mm $$times$$ 0.5 mm IIa single-crystal diamond) with a scanning microbeam was performed with using scanning focused 3 MeV proton microbeam. Photon counts of IBIL from the diamond were continuously observed and the changes were obtained during the process of PBW. An micro-IBIL image at a-band line of diamond crystal was successfully visualized as the PBW pattern on it through the monochromatic photon counting system of IBIL.

The development of the fast single-ion hit system was continued from last year in order to improve the quality of microbeam and single-ion hit at the TIARA cyclotron of the Japan Atomic Energy Agency. The major subjects were reduction of miss hit, no hit on the targeted point by any ions in the single-ion-hit irradiation, and development of a real-time detection system of single-ion hit. A new microbeam of 400 MeV-Fe with a spot diameter of about 4 $$mu$$m was also developed and supplied to an experiment.
