The F54L mutation of Thioredoxin shows protein instability and increased fluctuations of the catalytic center

Neuronal degeneration and chronic kidney disease have been observed in Trx-F54L mutant rats; however, the details of why the Trx mutation causes these phenomena remain unknown. This study investigates molecular changes caused by the F54L mutation and explores the factors contributing to the Adem rat phenotype. Results showed that the F54L mutant was less active than WT Txn1 on insulin substrates, but no significant difference in redox potential was observed. On the other hand, F54L is more sensitive to degeneration in the body temperature range than the wild type. Furthermore, the crystal structure showed that F54 formed hydrophobic interactions with the surrounding hydrophobic amino acids. Moreover, the MD simulation results show increased fluctuations around the F54L mutation, as well as a tendency for the distance between residues C32 and C35 at the catalytic center to widen. The molecular dynamics of the F54L mutation may affect the substrate reduction activity, thermal stability, and kinetic stability.

We investigate the effect of easy-axis metamagnetic crossover on superconductivity in UTe$$_2$$ along the $$a$$-axis through measurements of AC susceptibility, magnetization, and the magnetocaloric effect. In ultra-clean single crystals, we identify a field-induced phase transition within the superconducting state at 5.6 T, driven by metamagnetism. This transition leads to a high-field superconducting state, significantly increasing the upper critical field to 12 T. A sudden increase in entropy at the transition suggests a self-reconstruction of the order parameter, enabling multi-component superconducting states to adapt to external perturbations.

Creep characteristic of the thick laminated rubber bearing made of natural rubber installed in the actual vibration isolation building for 55.2 years could be estimated by using existing measurement data and newly measurement data. Creep measurement data sets were examined by simple regression equation of recommended by JIS K6410-2. Regression analysis showed that the average creep displacement characteristics could be described by a power law. Average creep displacement of the thick laminated rubber bearings installed the building for 60 years was estimated 3.4 mm by the regression equation. The method of JIS K6410-2 is useful for forecasting creep displacement.

Based on the regulations (the safety regulation of Tokai Reprocessing Plant, the safety regulation of nuclear fuel material usage facilities, the radiation safety rule, the regulation about prevention from radiation hazards due to radioisotopes, which are related with the nuclear regulatory acts, the local agreement concerning with safety and environment conservation around nuclear facilities, the water pollution prevention act, and by law of Ibaraki Prefecture), the effluent control of liquid waste discharged from the Nuclear Fuel Cycle Engineering Laboratories of Japan Atomic Energy Agency has been performed. This report describes the effluent control results of the liquid waste in the fiscal year 2023. In this period, the concentrations and the quantities of the radioactivity in liquid waste discharged from the reprocessing plant, the plutonium fuel fabrication facilities, and the other nuclear fuel material usage facilities were much lower than the limits authorized by the above regulations.

To secure the seismic safety of the thin-walled mechanical components and piping under a severe design earthquake level, employing a three-dimensional (3D) seismic isolation system has been planned in a sodium-cooled fast reactor. The development results of the 3D isolation system have been reported in previous papers so far. Its update is reported in Part 7 to Part 9. Part 7 describes the overview of the development, the test plan of the isolation system in the assembled state of each element, and the performance of individual isolation elements. In part 8, the performance of the isolation device that each element was assembled into was investigated through loading tests. Part 9 reports analytical studies by an analysis model validated based on the insight of the test results.

The normal-conducting state of the superconductor UTe$$_2$$ is studied by entropy analysis for magnetic fields along the $$b$$-axis, obtained from magnetization using the relation $$(partial M/partial T)_B=(partial S/partial B)_T$$. We observe a strong increase in entropy with magnetic field due to metamagnetic fluctuations (spatially uniform, $$Q=0$$). The field dependence is well described by the Hertz-Millis-Moriya theory for quantum criticality of itinerant metamagnetism. Notably, the lower bound of the quantum-critical region coincides well with the position of the minimum in the superconducting transition temperature $$T_c(B)$$. Hence, our results suggest that $$Q=0$$ fluctuations reinforce the superconductivity.

The vortex dynamics in the spin-triplet superconductor, UTe$$_2$$, are studied by measuring the DC electrical resistivity with currents along the $$a$$-axis under magnetic fields along the $$b$$-axis. Surprisingly, we have discovered an island region of low critical current deep inside the superconducting (SC) state, well below the SC upper critical field, attributed to a weakening of vortex pinning. Notably, this region coincides with the recently proposed intermediate-field SC state. We discuss the possibility of nonsingular vortices in the intermediate state, where SC order parameter does not vanish entirely in the vortex cores due to the mixing of multiple SC components.
