Tumor radioresistance caused by radiation-induced changes of stem-like cell content and sub-lethal damage repair capability

Alive cancer cells after fractionated irradiations with 2 Gy X-rays per day for more than 30 days show clinically relevant radioresistant. Such radioresistance is experimentally interpreted to attributed to the increment of stem-like cell content. However, only an experimental approach cannot clarify the cell responses (DNA damage and cell death induction) of cancer stem cells, so the radioresistant mechanisms remain uncertain. In addition to the conventional cell experiments using radio-resistant cell lines established after fractionated irradiations, in this study we developed a mathematical model (so called integrated microdosimetric-kinetic (IMK) model) explicitly considering cancer stem-like cell content and DNA damage responses and investigated radioresistant mechanisms acquired after fractionated irradiations. The IMK model analysis suggested that the changes of stem-like cell fraction and DNA repair efficiency play important roles of radioresisitance acquired after irradiations. Considering these into the IMK model, we successfully reproduced the experimental survival of various cell lines and various irradiation conditions. This work would contribute to not only the precise understanding of the radioresistant mechanisms induced after irradiation but also predicting curative effects with high precision.

The proton linac installed in the Ibaraki Neutron Medical Research Center is used for production of the intense neutron flux for the Boron Neutron Capture Therapy (BNCT). The linac consists of the 3-MeV RFQ and the 8-MeV DTL. Design average beam current is 10mA. Target is made of Beryllium. First neutron production from the Beryllium target was observed at the end of 2015 with the low intensity beam as a demonstration. After the observation of neutron production, a lot of improvement s was carried out in order to increase the proton beam intensity for the real beam commissioning. The beam commissioning has been started on May 2016. The status of the commissioning is summarized in this report.

The installation of Cs-seeded RF-driven H$$^{-}$$ ion source (RF ion source) to J-PARC LINAC is scheduled in 2014. Similarly the replacement of RFQ III from RFQ is scheduled in 2014. The test stand of the cooperation of RF ion source & RFQ III is being made in the J-PARC LINAC building for the beam acceleration examination now. The J-PARC control group designed test stand control system by thinking that test stand control system had to be equal to J-PARC accelerator control system. Specifically, Introduction of MPS for protect an apparatus. Implementation of EPICS environment for remotely controlling the equipment. It is construction of the timing system for sending a timing signal to each accelerator component device. This report describes construction of the control system in a test stand.

Beam loss reduction by the beam duct realignment in the J-PARC linac beam transport line

In the J-PARC linac, almost all the cavities and magnets have been precisely aligned because the accelerator tunnel has been deformed by the 2011 Tohoku Earthquake. In the beam transport line called MEBT2 and A0BT at the downstream of the drift tube linac, the beam duct has been roughly aligned after the precise alignment of the quadruple doublets. During the first beam operation after the earthquake, remarkable beam loss and residual radiation have been recognized at the MEBT2 and A0BT. As the result of the duct position measurement, the misalignement including over 10 mm shift from the beam axis was found. By conducting the beam duct realignment, the beam loss and the residual radiation were successfully decreased. In this paper, the procedure of the beam duct alignment and the beam loss due to the misalignment are described.

Surface preparation and characterization of single crystalline $$beta$$-FeSi$$_{2}$$

Well-defined clean surfaces of single crystalline $$beta$$-FeSi$$_{2}$$ have for the first time been prepared and characterized at the atomic scale. Surface oxide was removed by heating the crystal at 850 $$^{circ}$$C in ultra-high vacuum (UHV), resulted in an atomically-flat, clean surface. Scanning tunneling microscope (STM) and low-energy electron diffraction (LEED) measurements reveal the absence of long-range surface reconstruction in the low-index (100), (101) and (110) surfaces, which is unique among compound semiconductors. However a significant number of surface defects could still be found within the clean surface. Characterizing and controlling of those surface defects will be necessary in order to use this material in applications.

Present status of JAEA-Tokai tandem accelerator

In FY2006, a tandem accelerator was operated for 201days. The maximum voltage was 18 MV and the accelerated ion species was 19 elements (26 nuclides). As the trouble case which occurred previous year, the change of the beam shape occurred often in the injection beam line of the tandem accelerator. The cause was the contact failure of the pole connector of the electrostatics quadrupole lens. Generally, it was difficult to confirm the connection status of the opened pole but we specified a connection abnormal part by the capacitive coupling method. This technique is useful technology which can be applied to the other electrostatics optics elements. Because the charging electric current flowed only about usual about 60% and the terminal voltage became unstable, we opened a tank and checked the charging system. As a result, a problem was confirmed to the damaging of resistance and the installation of the cable. In the seminar, the operation, maintenance and the accelerator development of the tandem accelerator in FY2006 are described.

In order to sustain safe and stable operation of JRR-3, it is necessary to measure and indicate appropriately the process values such as flow rate of coolant with the process instrumentation facilities of JRR-3. Whenever reactor facilities such as cooling pump or measuring device are maintained or refurbished, the process instrumentation facilities are calibrated with the appropriate criteria established by considering overall accuracy of the facilities. In this report, for all of the safety protection system, a part of the process instrumentation facilities, accuracy of each component and overall accuracy of the system are compiled. Using these date, the process instrumentation facilities would be maintained more effectively and objectively.
