"Live-autoradiography" technique reveals genetic variation in the rate of Fe uptake by barley cultivars

Genetic diversity in the rate of Fe uptake by plants has not been broadly surveyed among plant species or genotypes, although plants have developed various Fe acquisition mechanisms. We adopted the "Live-autoradiography" technique with radioactive $$^{59}$$Fe to directly evaluate the uptake rate of Fe by barley cultivars from a nutrient solution containing a very low concentration of Fe. Our observations revealed that the ability to acquire Fe from the low Fe solution was not always the sole determinant of tolerance to Fe deficiency among the barley genotypes.

The impacts of oxygen in rhizosphere on the symbiotic nitrogen fixation in real-time was evaluated under various O$$_{2}$$ partial pressure. Soybean nodules were treated with mixed gas containing N$$_{2}$$ labeled with a radioisotope $$^{13}$$N and various concentrations of O$$_{2}$$, and the nitrogen fixation activity in the nodules was visualized by the positron-emitting tracer imaging system (PETIS). The results showed that under normal condition the nitrogen fixation ability of soybean plant was higher compared to that of under the other conditions (0% O$$_{2}$$ and 10% O$$_{2}$$). There was no significant difference in the nitrogen fixation activity between 0% O$$_{2}$$ and 10% O$$_{2}$$.

We investigated how volcanic CO$$_{2}$$ emission affects the $$^{14}$$C concentrations in tree rings of beech trees in the Hakkoda Caldera in Aomori, which is one of high emission regions of volcanic CO$$_{2}$$ in the world. There was a tendency that the obtained $$^{14}$$C concentrations were, surprisingly, quite near to the values of the concurrent atmospheric CO$$_{2}$$ and decreased from 1991 to 2001. The $$^{14}$$C concentrations in caldera are lower than those of the control samples taken from a beech living outside of the caldera. We revealed the possibility that we can solve the local dynamics of fossil CO$$_{2}$$ emission using $$^{14}$$C in tree rings. However, the maximum difference of $$^{14}$$C between inside and outside of the caldera was 14 permil. This result showed that the volcanic CO$$_{2}$$ mixed with atmospheric CO$$_{2}$$ by 4.4 ppm or less. As the trunk absorbing CO$$_{2}$$ exist in height that is several meters higher than the volcanic CO$$_{2}$$ vents, the low mixing may occur.

Imaging of the nodule function in leguminous plants who utilize nitrogen gas in the air as nutrition

The nodule is a symbiotic organ developed on roots of leguminous plants which have been infected with rhizobia, a group of soil bacteria. The plants with nodules can obtain nitrogen nutrition as ammonia converted from nitrogen gas in the air by rhizobia in the nodules. This function, "symbiotic nitrogen fixation", plays a fundamental role in the food production in the world. Recently, we reported that we have successfully visualized the symbiotic nitrogen fixation in soybean plants non-invasively using a positron-emitting tracer imaging system (PETIS) and nitrogen-13-labelled nitrogen gas tracer. In this article, this study is described with the scientific and social backgrounds.

The accelerator served mainly for graft-polymerization for new material development, radiation effect study on semiconductors, and various experiments of outside users. The annual operation time for the electron accelerator is 597.4h(517h for vertical beams, 80.4h for horizontal beams) The Co-60 $$gamma$$-ray irradiation facilities consist of three buildings (8 rooms) and cover a wide dose-rate range from 0.04Gy/h to 20kGy/h with eight irradiation cells. The annual operation time for the cobalt first, second irradiation facilities and food irradiation facility is 20,844 h, 6,335 h and 3,150 h.
