Increase in cell motility by carbon ion irradiation via the Rho signaling pathway and its inhibition by the ROCK inhibitor Y-27632 in lung adenocarcinoma A549 cells

The effect of carbon ion irradiation on cell motility through the Rho signaling pathway in the human lung adenocarcinoma cell line A549 was studied. At 48 h after irradiation, the cell motility of A549 cells became significantly greater, and the formation of protrusions significantly increased in cells irradiated with carbon ion. The observed increase in cell motility due to carbon ion irradiation was similar to that observed due to X-ray irradiation. Western-blot analysis showed that carbon ion irradiation increased P-MLC2-S19 expression compared with in unirradiated controls, while total MLC2 expression was unchanged. Exposure to a non-toxic concentration of Y-27632, a specific inhibitor of ROCK, reduced the expression of P-MLC2-S19 after C-ion irradiation, resulting in a significant reduction in migration. These data suggest that carbon irradiation increases cell motility in A549 cells via the Rho signaling pathway and that ROCK inhibition reduces that effect.

The electronic structure of spinel-type chalcogenide CuIr$$_{2}$$S$$_{4}$$, which exhibits a metal-insulator transition (MIT) as a function of temperature, has been studied by photoemission spectroscopy (PES). Below the transition temperature $$T_{mathrm{MI}}$$, the valence-band PES spectra indicate a gap formation, while the overall valence-band structure is shifted to the higher binding-energy side. The line shape of the Ir 4${it f}$ core-level PES spectra shows a dramatic change across $$T_{mathrm{MI}}$$, which may be associated with the variation of the Ir 5${it d}$ electron states.

Intravascular brachytherapy is a novel therapy for preventing the restenosis of coronary artery by use of low-dose irradiation. JAERI and Kyoto University have been developing $$^{192}$$Ir radiation sources by the cooperative research project entitled as "The research on safety and effectiveness of the intravascular brachytherapy for preventing restenosis of the coronary artery disease" since 1998. The radiation source was introduced into the stenosis through a catheter (a guide-tube to insert directly into vascular) to irradiate the diseased part. Ten $$^{192}$$Ir seed sources ($$phi$$0.4 mm $$times$$ 2.5 mm) were positioned between nylon spacers ($$phi$$0.3 mm $$times$$ 1.0 mm) in a flexible covering tube and the tube was plugged with a core-wire; the tube was shrunk to fix the inside materials and the size is 0.46 mm in diameter and 3 m in length. The physically optimal design was determined to insert the radiation source easily into vascular and to get the dose uniformity in the diseased part. The production method of the radiation source, which is practical to use in the clinics was also established.

Effect of hydrogen and helium irradiation on optical property of tungsten

Polycrystalline tungsten samples have been irradiated by 19 keV pulsed hydrogen and helium beam up to 1$$times$$10$$^{24}$$/m$$^{2}$$. Optical reflectivity has been measured with an ultraviolet-visible and near-infrared spectrophotometer, and a Fourier transform-infrared (FT-IR) spectrometer for the wave length between 190 - 2400 nm, 1.4 - 20 $$mu$$m, respectively. In addition, optical constants have been calculated by results measured using a spectroscopic ellipsometer. Surface morphology has been also examined with a scanning electron microscope. Loss of metallic gloss and remarkable reduction of reflectivity (190-2400 nm) occurs by the hydrogen and helium irradiation of $$sim$$1022 He$$^{+}$$/m$$^{2}$$. In particular, helium beam irradiation at a peak temperature 2600 $$^{circ}$$C causes remarkably decrease of the reflectivity due to fine uneven morphology and bubble formation near-surface. The reduction of the reflectivity is considered to be mainly due to the porous structure with nano-scale hydrogen and helium bubbles.

This study aimed to investigate the effect of carbon ion (C-ion) irradiation on cell motility through the ras homolog gene family member (Rho) signaling pathway in the human lung adenocarcinoma cell line A549. Cell motility was assessed by a wound-healing assay, and the formation of cell protrusions was evaluated by F-actin staining. Cell viability was examined by the WST-1 assay. The expression of myosin light chain 2 (MLC2) and the phosphorylation of MLC2 at Ser19 (P MLC2-S19) were analyzed by Western blot. The data suggest that C-ion irradiation increases cell motility in A549 cells via the Rho signaling pathway and that ROCK inhibition reduces that effect.
