Times Cited Count:4 Percentile:47.6(Multidisciplinary Sciences)Hyaluronan synthesis inhibitor 4-methylumbelliferone (4-MU) is a candidate of radiosensitizers in X-ray therapy. The curative effects under such 4-MU administration have been investigated in vitro; however, the radiosensitizing mechanisms remain unclear. Here, we investigated the radiosensitizing effects under 4-MU treatment from cell experiments and model estimations. We generated experimental surviving fractions of human fibrosarcoma cells (HT1080) after 4-MU treatment combined with X-ray irradiation. Meanwhilst, we also modelled the pharmacological effects of 4-MU treatment and theoretically analyzed the synergetic effects between 4-MU treatment and X-ray irradiation. The results show that the enhancement of cell killing by 4-MU treatment is the greatest in the intermediate dose range of around 4 Gy, which indicates the involvement of intercellular communication. In addition, the oxidative stress level, which leads to DNA damage induction, significantly increased under 4-MU treatment, and the radiosensitization by 4-MU can be suppressed by the inhibitors for intercellular communication. These findings suggest that the synergetic effects between 4-MU treatment and irradiation are predominantly attributed to intercellular communication and provide more efficient tumour control than conventional X-ray therapy.

Times Cited Count:23 Percentile:81.57(Biology)In radiotherapy, it is recognized that cancer stem cells (CSCs) in tumor tissue shows radio-resistance. However, the relationship between content percentage of the CSCs and dose-response curve on cell survival remain unclear. In this study, we developed a stochastic model considering progeny cells and stem cells, and investigated the impact of stem cells on radio-sensitivity. From the flow-cytometric analysis (cell experiments), the content percentage of stem cells was 3.2% or less which agreed well with the model estimation from the cell survival curve. Based on the verification, it is suggested that cell survival in high-dose range is largely affected by the CSCs. In addition, regarding the sub-population of stem cells, the present model well reproduces the dose response on lethal lesions to DNA comparing with the conventional LQ model. This outcome indicates that the stem cells must be considered for describing the dose-response curve in wide dose range.

Times Cited Count:3 Percentile:72.17(Physics, Multidisciplinary)We report the design of the beam-transport system (especially the vertical geometry) for a cold-neutron disk-chopper spectrometer AMATERAS at J-PARC. Based on the elliptical shape, which is one of the most effective geometries for a ballistic mirror, the design was optimized to obtain, at the sample position, a neutron beam with high flux without serious degrading in divergence and spacial homogeneity within the boundary conditions required from actual spectrometer construction. The optimum focal point was examined. An ideal elliptical shape was modified to reduce its height without serious loss of transmission. The final result was adapted to the construction requirements of AMATERAS. Although the ideas studied in this paper are considered for the AMATERAS case, they can be useful also to other spectrometers in similar situations.

Advanced technique has been also realized by taking an ability of event-recording system altogether with an improvement of a chopper system, realizing multi-Ei method. Tritium concentrations of river water, precipitation and ground water around the NIFS site were determined by low background liquid scintillation measurement system combined with the electrolysis using solid polymer electrolyte. The electric conductivity and flow rate of the river and isotopic ratio of oxygen and hydrogen of water samples were also measured. The tritium concentrations in precipitation showed the seasonal variation and the range were 0.09-0.78 Bq/L. The tritium concentrations of river water and ground water were almost constant, 0.34 and 0.24 Bq/L respectively. The simple dynamic model for the site around the NIFS facilities was developed using measured data, and the behavior of tritium was simulated.

The construction of main part of the spectrometer has been completed in spring of 2009. Soon after that, we have started the commissioning work on AMATERAS. The performance of AMATERAS has been examined by test experiments in the course of commissioning. In parallel to these works, we have started the user program on AMATERAS from December 2009 and we are getting scientific results from our spectrometer. In this presentation, we will report the current status of AMATERAS including the results of performance tests and some of examples of scientific outputs.

Times Cited Count:27 Percentile:58.17(Geochemistry & Geophysics)Patterns of the entire range of REE in crude oils and coexisting water, collected from mud volcanoes in Xinjiang Province of China, are reported here for the first time. Crude oils show light REE enriched patterns with flat or depleted patterns in heavy REE, when normalized to chondrite. The REE concentrations in crude oils are larger than those in coexisting water by a factor of more than one hundred. Considering the hydrophobicity of oil and the high ionic characteristics of REE, it is strongly suggested that REE form complexes with ligands present in the crude oils. Based on the C NMR spectroscopy, it is found that small amounts of phenol and carboxyl groups are contained in the crude oil samples, which could possibly provide complexing sites for REE. REE patterns of crude oils are similar to those of coexisting mud samples collected from the same mud volcanoes, which suggests that the REE in crude oils are derived from rocks and sediments where crude oils were generated.

Future synchrotron light source using a 5-GeV energy recovery linac (ERL) is under proposal by our Japanese collaboration team, and we are conducting R&D efforts for that. We are developing high-brightness DC photocathode guns, two types of cryomodules for both injector and main superconducting (SC) linacs, and 1.3 GHz high CW-power RF sources. We are also constructing the Compact ERL (cERL) for demonstrating the recirculation of low-emittance, high-current beams using above-mentioned critical technologies.

"R&D Project on Irradiation Damage Management Technology for Structural Materials of Long-life Nuclear Plant" was carried out from FY2006 in a fund of a trust enterprise of the Ministry of Education, Culture, Sports, Science and Technology. The coupled irradiations or single irradiation by JOYO fast reactor and JRR-3 thermal reactor were performed for about two years. The irradiation specimens are very important materials to establish of "Evaluation of Irradiation Damage Indicator" in this research. For the acquisition of the examination specimens irradiated by the JOYO and JRR-3, we summarized about the overall plan, the work process and the results for the study to utilize these reactors and some facilities of hot laboratory (WASTEF, JMTR-HL, MMF and FMF) of the Oarai Research-and-Development Center and the Nuclear Science Research Institute in the Japan Atomic Energy Agency.

The e in the ILs have same absorption peak at 1100 nm with a molar absorption coefficient of 1.5-2.310 dmmolcm. The reaction rate constant of e with Pyrene (Py) was 1.5-3.510 dmmols. These values were about one order of magnitude higher than that of diffusion controlled limit, which was calculated from measured viscosity. The dry electron (e) reacts with Py, and its rate constant in DEMMA-TFSI was 7.910 dmmols, three orders of magnitude higher than that of the e reactions.

The concept of "Waste Management System" which manages all the waste data from generation to disposal in Japan Atomic Energy Agency (JAEA) was studied. At first, development policy was defined on investigation results of the present state and problem of waste management in JAEA, and target information supplied by "Waste Management System" were extracted. Then over 400 data items to be managed in this system, system configuration and specific functions were shown as system concept. The waste data managed on this system are classified into two categories; for quality assurance of waste packages and for optimization study of waste conditioning and disposal and information service. This system consists of analytical part (i.e. program) and management part (i.e. database) for each data, then all these parts are constructed on network.
