Development of "leaky" liposome triggered by radiation applicable to a drug reservoir and a simple radiation dosimeter

Effects of lipid compositions and the content ratios on the membrane permeability of liposome have been investigated using soft X-rays as ionizing radiation (IR) sources. Three-component liposome constituted from an unsaturated phosphatidyl choline (PC), a saturated PC, and cholesterol was selected in a series of the study. As the results, following characteristics of IR-sensitive liposome were discovered: (1) A PC with bis-allylic hydrogen is essential as a component of unsaturated PC., (2) Dilinoleoyl PC (DLOPC) is optimal as a component of unsaturated PC., (3) Either distearoyl PC (DSPC) or dipalmitoyl PC (DPPC) is optimal as a component of saturated PC., (4) Lower dose- rate makes the liposome more IR-sensitive., (5) Lower concentration of liposome makes the liposome more IR-sensitive., (6) There is a region for optimal DLOPC content ratio for the IR-sensitive liposome around 5 $$sim$$ 20 mol%., and (7) There is a region for optimal cholesterol content ratio for preparing a stable liposome around 30 $$sim$$ 60 mol%. These fundamental laws for IR-sensitive liposome will contribute to understanding of biomembrane damage by IRs and developing a new-conceptual pharmaceutical available in radiotherapies.

Real-time X-ray observation of solidification from undercooled Si melt

Grain refined microstructure is often obtained spontaneously in the solidification of metals and semiconductors from the undercooled melt without any external forces. Although it has been reported that the grain refinement is mainly caused by the fragmentation of the dendrites, the dynamic process of the fragmentation of dendrites has not been fully understood because the microstructure after the solidification has been analyzed. Here, we present a time-resolved 2- dimensional X-ray diffraction experiment on the solidification of Si from the undercooled melt. The number of diffraction spots observed at low undercoolings ($$Delta T<$$100K) did not increase at the plateau stage, while the diffraction pattern at medium undercoolings (100K$$<Delta T<$$200K) changed from the spots with the tail to rings with the lapse of time. Both this result and high speed video imaging suggested that the high-order arms of the dendrites mostly detached from the main stems because nucleation could not be expected at the melting point after recalescence. The several spots observed at low undercoolings drastically changed to rings at high undercoolings ($$Delta T>$$200K), which indicated the complete fragmentation of dendrite main stem as well as high-order arms. This complete fragmentation resulted in the grain refined microstructure.

Precise Measurement of density and structure of undercooled molten silicon by using synchrotron radiation combined with electromagnetic levitation technique

X-ray diffraction and density measurements have been simultaneously performed to investigate the atomic structure of molten silicon in wide temperature range including undercooling region by using the electromagnetic levitation technique. The density was obtained from the mass and the shape of levitated sample by non-contact method based on the image analysis technique. X-ray diffraction experiments were performed by using the synchrotron radiation at SPring8, Japan. From structural analysis of undercooled molten silicon, first nearest neighbour coordination numbers and interatomic distances were about 5 and 2.48$$AA$$ with no dependence on temperature in the range of 1900-1550 K. We conclude as a result that the short-range order based on tetrahedral bonds of undercooled molten silicon does not change with the degree of undercoolings but medium-range order changes by the degree of undercoolings.

Development of ionizing radiation-sensitive liposome for radio-chemo-concurrent cancer therapy

We have developed a ionizing radiation-sensitive liposome aiming to construct a release control system which the incorporated anticancer agents in liposomes accumulated at tumor can be released following to destruction of them by the irradiation. We predict that one of most effective chemical species injuring liposome is hydroxyl radical produced by radiolysis of water. According to the hypothesis, as the first step of the development, we have investigated the relationship between lipid compositions in liposome (a 3-membered system: cholesterol, a saturated phospholipid, and an unsaturated phospholipid) and radiation sensitivity, using a soft X-ray equipment (Softex M-150WE). Based on the results from the X-ray experiments, a liposome with both a boron compound and anticancer agent was prepared, considering its use for boron neutron capture therapy.

Chronological and geochemical studies of calcium carbonates can provide precious information for changes in geochemical condition in deep geological environments. Because the carbonate can be found as common filling minerals in rocks, age zoning and spatial distribution of chemical composition in the carbonate could be a wide-use indicator to reconstruct the past environmental changes. Calcite and other carbonate materials have been dated by Isotope Dilution-Thermal Ionization Mass Spectrometry. On the other hand, a few of U-Pb dating studies in a micro scale area (less than 10 micrometer) such as using laser ablation-multiple collector-inductively coupled plasma mass spectrometry (LA-MC-ICPMS) has been conducted on the carbonate. The U-Pb dating technique for the carbonates using LA-MC-ICPMS is still immature partly because of a lack of consensus international carbonate reference materials for the in-situ dating technique. In order to solve this problem, we performed a survey on the carbonate reference materials that may be suitable for high precision U-Pb dating using LA-MC-ICPMS. Additionally, we conducted experiments to make compositionally homogeneous carbonate (calcite) reference materials.

We detected weak infrared absorption by H$$_{2}$$ trapped and polarized in solid CO$$_{2}$$ and evaluated the strength of the local electric field just in the solid. By analyzing the time evolution of the spectrum, we determined the conversion rates of H$$_{2}$$ to examine the effect of the electric field on the conversion.

Molecular clouds are composed of gases and interstellar dust. The dust is covered with an ice mantle containing a variety of molecules such as H$$_2$$O and CO and is exposed to ultraviolet radiation. In the present study, we investigated the photochemical reaction in the ice mantle utilizing isotopologues.

Nano-sized molecular clusters have been intensively investigated in a spectroscopic way to reveal the nature of intermolecular interaction. Compared with the infrared (IR) range, only a few spectral measurements in the terahertz (THz) range have been reported, so that the assignment of the vibrational mode to THz absorption was not established even for the water dimer. In the present study, we constructed an experimental apparatus for THz-IR absorption spectroscopy in an ultrahigh vacuum and measured spectra of deuterated-water clusters formed in solid Ar to identify the THz absorption peaks of the dimer, trimer, and tetramer.
