Times Cited Count:28 Percentile:80.14(Agronomy)A few carnation cultivars are known to display a peculiar dusky color supposedly caused by anthocyanic vacuolar inclusions (AVIs). The hereditary pattern suggests that the peculiar color is controlled by a single recessive factor tightly linked with existence of AVIs containing non-acylated anthocyanins. To diversify the peculiar color carnation, we produced a bluish purple line displaying a highly novel metallic appearance by crossbreeding. By subjecting the line to ion-beam irradiation, we generated metallic reddish purple, metallic crimson and metallic red lines. All four metallic lines did not have transcripts for anthocyanin malyltransferase. In contrast to the dusky color types, metallic lines have highly condensed AVIs and water-clear vacuolar sap in the petal adaxial epidermal cells. We demonstrated that (1) a factor generating the AVIs is inactivated anthocyanin malyltransferase gene, (2) AVIs in water-clear vacuolar sap in the adaxial epidermal cells generate the novel metallic appearance, and (3) ion beam breeding is a useful tool for increasing metallic colors by changing anthocyanin structure and the level of AVIs.

Mutation breeding is sometimes perceived as a random process. In this review, we describe our recent progress in developing a more efficient mutagenesis technique using ion beam irradiation combined with sucrose pretreatment or subsequent re-irradiation. To shorten the time required for breeding new cultivars of cyclamen, we identified anthocyanin biosynthesis genes and examined the effectiveness of PCR screening of irradiated deletion-mutant candidates at early growth stages. We believe this research is a step toward more efficient and controlled mutation breeding using ion beams.

The flower colour of gra6 (wild-type) is pink-purple in the flower. On the other hand, flowers of gra50 (white-flowered variant) exhibit a white colour in its flower. In this study, the relationship between floral pigmentation and expression of several anthocyanin biosynthesis genes was investigated in gra6 and gra50. The pigments in the flower consist mainly of malvidin 3,5-diglucoside in gra6. White-flowered gra50 possessed lower amounts of anthocyanins, but higher amounts of flavonols compared to gra6. Gene expression analysis demonstrated that expression of the dihydroflavonol 4-reductase gene 2 () was lower in gra50 compared with gra6, whereas the expression of other key genes did not differ greatly. These results suggest that the white-flowered variant of gra50 may result from a defect in expression of the gene.

To clarify the mechanism of growth and development of the fruit of eggplant,COwas fed to a leaf and the translocation of C-labeled photoassimilate into the fruit was monitored by PETIS. Continuous images of the translocation of C-photoassimilate from the leaf to the fruit were successfully made, with a shorter time resolution than ever reported before. The PETIS images indicated that translocation of photoassimilate within the fruit was not uniform. The velocity of photoassimilate translocation in the peduncle and the changes in the rate of translocation of photoassimilate in the fruit were estimated by analysis of the PETIS data. The advantages of PETIS measurements in physiological studies of source sink relationships are discussed.
