Disruption of the gene causes hypersensitivity to Ultraviolet B light and -rays in Arabidopsis; Implication of the presence of a translesion synthesis mechanism in plants

われわれは高等植物における紫外線応答機構を解明する目的で、イオンビームを照射を行ったシロイヌナズナから紫外線感受性変異株を単離した。の幼植物体に対してUV-B照射を行うと、暗条件下及び明条件下において根の伸長が抑制されるという表現型を示した。の染色体構造を詳しく解析したところ、株では第一染色体が3か所で切断されて、生じた断片が転座や逆位をおこしており、これによって損傷トレランス機構に関与するとみられる遺伝子が破壊されていた。変異株はDNAの複製を阻害することが知られている線やMMCなどの処理に対しても高い感受性を示した。また、紫外線照射後の幼植物体の根にBrdUを取り込ませてDNA合成効率を調べると、rev3-1株では野生型に比べてDNA合成効率が低下していた。以上のことから、rev3-1では遺伝子の欠損のために、紫外線で生じた損傷によってDNA複製が停止し、その結果根の伸長が阻害されているという可能性が示唆された。
In order to investigate UV-response mechanisms in higher plants, we isolated a UV-sensitive mutant, , in . The root growth of was inhibited after UV-B irradiation under both photoreactivating and non-photoreactivating conditions. We found that chromosome 1 of was broken at least at three points, causing chromosome inversion and translocation. A gene disrupted by this rearrangement encoded the catalytic subunit of DNA polymerase (), which is thought to be involved in translesion synthesis. The seedlings were also sensitive to -rays and MMC, which are known to inhibit DNA replication. The incorporation of BrdU after UV-B irradiation was less in than in the wild type plant. These results suggest that UV-damaged DNA interrupted DNA replication in the mutant, leading to the inhibition of cell division and root elongation.
