Poplars (${it Populus}$ spp.), is favored for plantation forestry and often serving as a vegetative pioneer species for its ecological and economic importance. However, no information is available particularly on development of salt or drought tolerant mutants in poplar. Insufficient irradiation system for screening of such mutants might be one of the reasons for that. This research focused on screening of salt tolerant mutants by use of in vitro regeneration and radiation mutagenesis. Here, we are presenting a) direct regeneration system in ${it Populus}$ through in vitro culture, b) optimization of irradiation system for woody plant, and c) identification of the mutants that produces roots in medium with 75 mM NaCl. Hopefully, these mutants will fulfill our initial goal of improve woody plants through radiation mutagenesis.
