The detector system consists of high-purity germanium detectors with BGO Compton suppressors. The signals from the detectors are readout with a digital acquisition system. The absolute energy accuracy, energy and timing resolutions, photo-peak efficiency, the performance of the Compton suppressor, and high count rate durability are studied with standard -ray sources and in-beam experiment using Al()Si resonance reaction. The detection system was demonstrated at Paul Scherrer Institute. A calibration method for a photon detector at a muon facility using muonic X-rays of Au and Bi is proposed.

被引用回数:18 パーセンタイル:83.17(Chemistry, Physical)We demonstrate a high thermoelectric figure of merit (zT) of 1.45 at 900 K for Ge doped (4-10 mol%) -type PbS, which is the one of the highest values among all -type PbS-based thermoelectric materials. This high performance is achieved by simultaneous (a) enhancement of covalency in chemical bonding which increases the electrical conductivity, and (b) reduction of lattice thermal conductivity to an ultra-low value of 0.56 W mK at 900 K by the introduction of nanometer-sized (5-10 nm) precipitates of PbGeS in PbS matrix which strongly scatter the heat-carrying phonons. The presence of low-lying transverse acoustic (TA) and longitudinal acoustic (LA) phonon modes at 48.24 cm and 91.83 cm, respectively are experimentally revealed from inelastic neutron scattering (INS) experiments. The softening of low-frequency modes at a higher temperature and ultra-short phonon lifetime (1-4.5 ps) further explain the ultra-low thermal conductivity.

This experimental observable is obtained for the first time in the region around lead using an innovative approach based on inverse kinematics and the coincidence between the large acceptance magnetic spectrometer VAMOS++ and a new detection arm close to the target. The average fragment ratio and prompt neutron multiplicity are derived and compared with current knowledge from actinide fission. A striking consistency emerges, revealing the unexpected dominant role of the proton subsystem with atomic number between the = 28 and 50 magic numbers. The origin of nuclear charge polarization in fission and fragment deformation at scission are discussed.

The setup consisted of a collimation system producing a pencil beam with a thermal capture equivalent flux of about 10nscm at the target position and negligible neutron halo. The target was surrounded by an array of eight to ten anti-Compton shielded EXOGAM Clover detectors, four to six anti-Compton shielded large coaxial GASP detectors and two standard Clover detectors. For a part of the campaign the array was combined with 16 LaBr:(Ce) detectors from the FATIMA collaboration.

Species of the genus are ecologically important trees and often serving as a vegetative pioneer species because of their fast growth rates. The rapid growth of poplar is dependent on access to sufficient water in plantation sites and this is problematic. To overcome this problem, a forward genetics approach has been taken for developing salt/drought tolerant mutants by combination of radiation mutagenesis and in vitro regeneration in . Previously we screened regenerated shoots regenerated from stems exposed with cobalt-60 source of rays and isolated two mutant candidates named and . Only about 25% stem segments of both lines produced roots in RIM with 75 mM NaCl. The low frequency in root production in NaCl medium may be attributed to that these plants are chimeric. We also successfully obtained at least 2 salt tolerant candidates regenerated from stem segments irradiated with 320 MeV C ion beams.

Poplars are economically important trees that are favored for plantation forestry due to their capacity for rapid juvenile growth, which allows us to harvest them in a short span of time. However the rapid growth of poplar is dependent on access to plantation sites with sufficient water and this is problematic, particularly in less rainfall area. The patterns of episodic drought over the last decade suggest that the development of drought tolerant mutant plants could be a promising approach to increase sustained vegetation biomass and forest productivity. We developed a system for the regeneration of Lombardy poplar ( L. var. ) shoots from internodal stem explants. Using this system, shoots regenerated from 87% of the stem explants placed on Murashige and Skoog (MS) medium supplemented with 0.1 mg/L indole-3-acetic acid and 0.5 mg/L benzylaminopurine without undergoing callus formation. About 80% of the regenerated shoots developed roots on MS medium supplemented with 0.5 mg/L indole-3-butyric acid and 0.02 mg/L 1-naphthylacetic acid. The protocol presented here is simple and economical because it does not rely on pre-incubation in callus induction medium or repeated subculture in shoot induction medium containing -zeatin, an expensive substance. The regeneration system presented here could be used for evaluation of radiation sensitivity for Lombardy poplar tissues.

Deletion analysis and green fluorescent protein (GFP) / glutathione S-transferase (GST) pull-down assays showed that SMAP1 physically interacts with the CONSTITUTIVE PHOTOMORPHOGENIC-9 SIGNALOSOME (CSN) via the SMAP1 F/D-region. The extremely dwarf phenotype of double mutant confirms the functional role of in plant growth and development under limiting CSN functionality. Our findings suggest that SMAP1 is involved in the auxin response and possibly in other cullin-RING ubiquitin ligase (CRL)-regulated signaling processes via its interaction with components associated with related-to-ubiquitin (RUB) modification.

Today, aided mainly by genetics, researchers have identified the core complexes for auxin perception, including the auxin receptors TIR1/AFBs, SKP2 and ABP1. Forward chemical screening using auxin response markers has also played an important role in identifying small molecules affecting the auxin-signaling pathways and led to the screening of additional auxin related mutants. This approach has helped in identifying several unique molecular components of the auxin-signaling pathways, and discovering that would have been impossible using traditional screening of mutants against auxin. This review is focused on recently acquired knowledge of multiple auxin signaling pathways and related small compounds that can be used to dissect auxin signal transduction pathways. In addition, the involvement of distinct auxin-signaling machineries during gravity response is discussed.

被引用回数:35 パーセンタイル:60.82(Plant Sciences)2,4-Dは、IAAの化学的アナログで、オーキシンとして投与し植物の生長を制御する際に広く使用されており、IAAと共通した応答経路で作用すると考えられている。本論文では、2,4-Dに対して感受性が低下しているがIAAに対しては野生型と同様の感受性を示す変異体であるアンチオーキシン耐性変異体()について報告する。の2,4-D応答変化の特異性は、, などのマーカー遺伝子の発現や遺伝子のリアルタイムPCR法などにより確認された。2系統の変異遺伝子座を調べたところ、両者とも複数の遺伝子にまたがるゲノムDNAの欠失が起こっていた。そこで、相補試験、及びRNAi形質転換体の解析を行い、欠失領域に存在する単一の遺伝子が2,4-D応答変化に関与していることを証明した。この遺伝子は、機能未知のタンパク質をコードする遺伝子(:)で動植物には存在するが、菌類や原核生物には報告がないものであった。本研究により、SMAP1は2,4-D応答の調節因子であること、2,4-DとIAAに対する応答反応は部分的に異なる経路で引き起こされることが示唆された。

However, no information is available particularly on development of salt or drought tolerant mutants in poplar. Insufficient irradiation system for screening of such mutants might be one of the reasons for that. This research focused on screening of salt tolerant mutants by use of in vitro regeneration and radiation mutagenesis. Here, we are presenting a) direct regeneration system in through in vitro culture, b) optimization of irradiation system for woody plant, and c) identification of the mutants that produces roots in medium with 75 mM NaCl. Hopefully, these mutants will fulfill our initial goal of improve woody plants through radiation mutagenesis.

Development of suitable mutagenesis and screening system is necessary to isolate salt/drought tolerant mutants in hard wood plants, such as poplar. Here, we are introducing a specialized screening system with combination of regeneration and ion beam or ray irradiation. A high percentage of direct plant regeneration could be achieved from stem explants placed on Murashige and Skoog (MS) medium supplemented with indole-3-acetic acid and benzylaminopurine. To optimize dosage for mutagenesis, dose response relationship curve for shoot regeneration ratio had been established. Initial screening of salt/drought tolerance could be achieved by observing the ability of root development of regenerated shoots in 75 mM NaCl-supplemented rooting medium because wild-type regenerated shoots failed to develop roots in this medium. Hopefully, our system will help to reduce existing limitations of screening mutants in hard wood plants having a long life cycle.

For screening of drought/salt tolerant poplar mutants, regeneration system had been established. About 87% of the shoot explants produced regenerated shoots in shoot regeneration medium, whereas 77% regenerated shoots developed roots in root induction medium. Survival rate of the regenerated plants in soil was about 88%.Suitable dose of ion beams to screen mutants ranged between 2.5 Gy to 3 Gy for shoot explants in poplar. regenerated wild type poplar shoots did not develop any root system in rooting medium supplemented by 75 mM NaCl. Thus, the initial goal of this study to screen drought/salt tolerant mutants could be achieved by the use of well-established ion-beam facilities at Japan Atomic Energy Agency.

根の伸長試験により、合成オーキシン2,4-Dに耐性のある新規変異体()の分離に成功した。ボジショナルクローニングによりAAR3遺伝子は、DCN1タンパク質とPONYドメインを介して類似している核局在のタンパク質をコードしていることが明らかになった。本研究によりDCN1様タンパク質が根における2,4-D応答を制御していることが示唆された。

()遺伝子は、イオンビームで創出されたシロイヌナズナのアンチオーキシン抵抗性変異体の原因遺伝子であり、植物ホルモンの一種で、植物の発根や根の生長に作用する合成オーキシン2,4-Dの応答にかかわる因子として同定された遺伝子である。SMAP1がシロイヌナズナ抽出液中でタンパク質分解活性を制御するタンパク質複合体であるCOP9シグナロゾーム複合体(CSN)と結合していることが明らかにされている。そこで、今回、SMAP1とCSNとの関係について遺伝学的知見を得るために、変異体とCSNのサブユニットCSN5Aの変異体を交配し、二重変異体の作出を行った。二重変異体は、及びに比べ、胚軸が短いほか、子葉・本葉が小型化し、根の生長も遅いなどの形態的特徴が見られ、これらの特徴は、の導入で解消された。根における2,4-D感受性は、相対的に単独の変異よりも低下しており、生長した個体は著しくわい化し、稔性がほとんどなかった。以上の結果より、のみの変異体はほとんど形態的な特徴や生長異常は見られないが、CSNが十分に機能しない状態では、SMAP1が植物の正常な生長に重要な役割を果たしていることが示唆された。

われわれはこれまで、アンチオーキシンとして知られるPCIB(パラクロロイソ酪酸)を用いて根の伸長を指標に変異体のスクリーニングを行ってきた。その結果、少なくとも3種類の新奇変異体を得ることに成功した。その中の一つであるは、根の伸長試験において、PCIBのみならず2,4-Dにも感受性が低下した変異体であった。その原因遺伝子は、DUF298(Domain of Unknown Function 298)という機能不明のドメインを持つタンパク質をコードしていた。このタンパク質は核局在シグナルと思われる配列を持ち、プロトプラストを用いた一過的発現解析で、核に局在することが確認された。また、このタンパク質はDUF298領域を介してDCN-1(DEFFECTIVE IN CULLIN NEDDYLATION 1)タンパク質と相同性を有していた。しかし、オーキシン応答マーカーである及びを用いた2,4-D感受性試験では、野生型との間で明確な違いが観察されず、がこれまでに知られている作用機構とは異なる機構で2,4-Dの感受性に関与している可能性が示唆された。
