The AMS system is a versatile system based on a 5 MV tandem Pelletron type accelerator. The system has been used to measure carbon-14 (C), beryllium-10 (Be) and aluminium-26 (Al). In addition, the development of measurement of iodine-129 (I) has been started. The main use is measurement of C in geological samples for dating studies in neotectonics and hydrogeology. In order to increase the speed of sample preparation, we introduced the automated graphitization equipment and made a gas-strip line to collect dissolved inorganic carbon in groundwater samples. Measurement of Be and Al has been used for geoscience studies and the detection limit in the measurement of Be was improved by Be-counting suppression. Recently tuning of measurement condition of I has been progressed.

AMS is widely used for radiocarbon dating of geological samples. However, improvement in efficiency of sample preparation techniques are needed for high-time resolution dataset. In 2016, automated graphitization equipment (AGE3, IonPlus AG) has been installed in Toki Research Institute of Isotope Geology and Geochronology, Tono Geoscience Center, JAEA. Background values and carbon recovery rates during preparation process of AGE3 should be estimated before application in radiocarbon dating. In this study, the AGE3 system was evaluated using the international standard materials (IAEA-C1, C4, C5, C6, C7, C9 and NIST-SRM4990C) at JAEA-AMS-TONO. Graphite samples was prepared by the AGE3 system and radiocarbon concentration of these standards was measured by AMS. The results were agreement with the consensus values. Background values were 0.150.01 pMC (IAEA-C1) using the AGE3 system. Therefore, we concluded that the system can be adapted for radiocarbon dating of geological samples.

The AVF cyclotron was operated from May 9th in fiscal 2011 and all the planned experiments on April were cancelled due to scheduled blackouts originated from the Great East Japan Earthquake. The total operation time amounted to 3,038.4 hours. The extended operation from Friday evening to Saturday evening was carried out eight times in order to supply the cancellation on April. The regular yearly overhaul and maintenance were carried out.

Times Cited Count:72 Percentile:89.95(Plant Sciences)In order to identify the anthocyanin-related GST in cyclamen, four candidates of GSTs (CkmGST1 to CkmGST4) were isolated. Phylogenetic analysis indicated that CkmGST3 was closely related to PhAN9, an anthocyanin-related GST of petunia. Expression analysis at different developmental stages of petals revealed that CkmGST3 was strongly expressed in paler pigmented petals than in fully pigmented petals, in contrast to the constitutive expression of the other three candidates during petal development. This expression pattern of CkmGST3 was correlated with those of other anthocyaninbiosynthetic genes such as CkmF3'5'H and CkmDFR2. Molecular complementation of Arabidopsis , a knockout mutant of an anthocyanin-related GST gene, demonstrated that CkmGST3 could complement the anthocyanin-less phenotype of . Transgenic plants that expressed the other three CkmGSTs did not show anthocyanin accumulation. These results indicate CkmGST3 functions in anthocyanin accumulation in cyclamen.

Reduction of the beam changing time is required to increase the available beam time for users. We are developing a technique for reducing the changing time of the magnetic field of the cyclotron, since it takes a long changing time to form the magnetic field for acceleration. Since the deviation of the magnetic field from the desired value for acceleration in the start-up process of the magnetic field dose not decrease in a short time, a correction system of the cyclotron magnetic field with a bipolar power supply and a 5-turn coil was developed. We confirmed that this system enables the magnetic field to be controlled quickly with a pc-based PID controller.

The flower colour of gra6 (wild-type) is pink-purple in the flower. On the other hand, flowers of gra50 (white-flowered variant) exhibit a white colour in its flower. In this study, the relationship between floral pigmentation and expression of several anthocyanin biosynthesis genes was investigated in gra6 and gra50. The pigments in the flower consist mainly of malvidin 3,5-diglucoside in gra6. White-flowered gra50 possessed lower amounts of anthocyanins, but higher amounts of flavonols compared to gra6. Gene expression analysis demonstrated that expression of the dihydroflavonol 4-reductase gene 2 () was lower in gra50 compared with gra6, whereas the expression of other key genes did not differ greatly. These results suggest that the white-flowered variant of gra50 may result from a defect in expression of the gene.

The ratio of the beam changing time, which is the time for changing the operation condition of the cyclotron, to the whole operation time is large at the JAEA AVF cyclotron, since most irradiation time is short and frequent change of beams is required by users, mainly in materials science and biotechnology. In order to increase the efficiency of the use of the cyclotron, we are developing a technique for reducing the beam changing time, which is dominated by the start-up process of the magnetic field. A quick start-up process for the main magnetic field with a damping oscillation excitation allows to reduce the start-up time of the magnetic field.

High-frequency change of beams accelerated for users is required at the JAEA AVF cyclotron, since the irradiation time is short in most cases and a large variety of beams is used. The ratio of the beam changing time to the whole operation time is not negligible and the offering machine time for users is half the required one. We are developing a technique for reducing the changing time of the magnetic field of the cyclotron, which dominates the beam changing time. Since a precise and stable magnetic field is required for acceleration in the cyclotron, especially for advanced beam applications, such as microbeam production, a magnetic field measurement system with a nuclear magnetic resonance (NMR) magnetometer was developed to monitor a wide range of magnetic field for acceleration of variety of beams in the JAEA AVF cyclotron.

Some of the M2 plants derived from self-pollination of M1 plants irradiated at 2 Gy were flower-color mutants that retained desirable flower shape, flower size, and leaf color. One of the mutants bloomed novel red-purple flowers, the major anthocyanin of which was delphinidin 3,5-diglucoside. Because the major anthocyanins in flowers of spp. were previously restricted to malvidin, peonidin, and cyanidin types, the generation of a cyclamen containing mostly the delphinidin-type anthocyanin is an important breakthrough in cyclamen breeding. We expect this mutant to become not only a commercial cultivar itself, but also a valuable genetic resource for cyclamen breeding.
