Extraction of cytochrome c by a functionalized ionic liquid containing a crown ether

クラウンエーテルを有する機能性イオン液体へのシトクロムcの抽出

クラウンエーテルを分子内に有する機能性イオン液体[18C6mim][PF$$_{6}$$]を用いてタンパク質であるシトクロムcの抽出を行った。機能性イオン液体[18C6mim][PF$$_{6}$$]は水酸基を有するイオン液体[C$$_{2}$$OHmim][Tf$$_{2}$$N]あるいはエチル基を有するイオン液体[C$$_{2}$$mim][Tf$$_{2}$$N]に希釈して用いた。その結果、シトクロムcは水相から[18C6mim][PF6]を含んだ[C$$_{2}$$OHmim][Tf$$_{2}$$N]へ容易に抽出されることが明らかとなった。しかし、[C$$_{2}$$mim][Tf$$_{2}$$N]への抽出率は小さかった。また、[18C6mim][PF$$_{6}$$]は抽出剤dicyclohexano-18-crown-6 (DCH18C6)より抽出能は低いが、逆抽出操作においては大きな利点を示した。抽出における温度依存性を検討したところ、[18C6mim][PF$$_{6}$$]の濃度が小さい領域では大きく抽出温度に影響した。今回使用した機能性イオン液体[18C6mim][PF$$_{6}$$]は従来のイオン液体に比べ高い抽出能を有することが明らかとなった。

We carried out the extraction of a protein cytochrome c (Cyt-c) with an ionic liquid with an attached crown ether moiety in the imidazolium cation. The functional ionic liquid, [18C6mim][PF$$_{6}$$], was dissolved in two different ionic liquids as diluents, one having a hydroxyethyl group, [C$$_{2}$$OHmim][Tf$$_{2}$$N], and the other an ethyl group, [C$$_{2}$$mim][Tf$$_{2}$$N], respectively. The transfer of Cyt-c from the aqueous phase to the ionic liquid phase took place with [18C6mim][PF$$_{6}$$] in the [C$$_{2}$$OHmim][Tf$$_{2}$$N] system. Although the extraction ability was lower than that of dicyclohexano-18-crown-6 (DCH18C6), the functionalized ionic liquid system gave a better stripping performance than the ordinary crown ether system. The extraction efficiency was not influenced by operating temperature when the concentration of [18C6mim] was high whereas it was greatly affected by temperature at low [18C6mim] concentration.
