放射線抵抗性細菌デイノコッカス・ラジオデュランスのDNA2本鎖切断(DSB)修復機構におけるRecFOR経路の役割を明らかにするために、遺伝子破壊株を作製し、線に対する感受性を調べるとともに、切断されたゲノムDNAの修復過程,照射前後でのDNA修復タンパク質の細胞内量変動を解析した。野生株に比べて、遺伝子破壊株は線に対して高い感受性を示した。線照射後の切断されたDNA断片の修復過程を解析したところ、効率的な修復が見られず、RecFタンパク質がDSB修復機構に非常に重要な役割を担っていることを明らかにした。また、放射線照射後、遺伝子破壊株では、野生株と同様にRecAタンパク質の細胞内発現量が増加していたが、DNA組換え修復に必要なRecAタンパク質の活性化が起こらないことから、RecAタンパク質の活性化機構にRecFタンパク質が寄与していると考えられた。以上の結果から、ラジオデュランスのDSB修復機構において、RecFタンパク質はRecAタンパク質の活性化機構を介して非常に重要な役割を担ってることが示唆された。
To investigate the role of the RecFOR pathway in DNA double-strand break (DSB) repair in , we generated a disruptant strain and characterized the disruption effect. The disruptant strain exhibited hypersensitivity to rays than the wild-type strain. PFGE analysis showed that genome restitution ability following irradiation was severely impaired in the disruptant strain, suggesting that RecF has a crucial role in DSB repair. Furthermore, following irradiation, induction of RecA, which is essential protein for DNA recombination repair, was observed in the disruptant strain just as the wild-type strain. However a large amount of RecA protein in the disruptant strain remained inactive following irradiation. We suppose that the defect in RecA activation resulted in hypersensitivity to ionizing radiation and failure to repair radiation-induced DSB in the disruptant strain.
