イオン液体により蒸着した微生物起源ナノ粒子の電子顕微鏡観察

微生物細胞表面に生成したナノ粒子を走査型電子顕微鏡(SEM)で観察する方法としてイオン液体による蒸着法を検討した。実験では、親水性及び疎水性イオン液体を用いた。その結果、親水性イオン液体で蒸着した試料では、微生物細胞の形状を保ったままでナノ粒子を観察できた。一方、疎水性イオン液体を用いた場合には細胞がつぶれた形状を示した。この結果から、親水性イオン液体蒸着法は微生物起源のナノ粒子を観察する有効な方法であることがわかった。

We have observed biogenic nano-particles developed on cell surface of microorganism coated with ionic liquid by SEM. In the sample preparation, the microbial cells were coated with hydrophilic and hydrophobic ionic liquids. The biogenic nano-particles were clearly observed by SEM, when the sample were coated with hydrophilic ionic liquid. On the contrary, the cells were collapsed during SEM observation, when the sample were coated with hydrophobic ionic liquid. These results showed that the coating with hydrophilic ionic liquid is effective for analyzing nano-particles developed on microbial cell surface by SEM.
