デイノコッカス・ラジオデュランスの放射線耐性は、効率的なDNA2本鎖切断修復によるものである。ラジオデュランス由来のDNA修復欠損変異株KH311の解析から、ラジオデュランスの放射線耐性にかかわる新規の放射線誘導性遺伝子(と命名)を同定した。この遺伝子産物PprAは、DNA2本鎖切断部位に結合して、大腸菌エキソヌクレアーゼIIIからDNAを保護し、さらにATP依存性及びNAD依存性DNAリガーゼによるDNA末端結合反応を促進する活性を持つ。これらの結果から、ラジオデュランスがPprAを主要因子とする放射線誘導性の非相同性DNA末端結合による修復機構を持っていることが示唆された。
The extraordinary radiation resistance of results from the efficient capacity of the bacterium to repair DNA double-strand breaks. By analyzing the DNA damage repair-deficient mutant, KH311, a unique radiation-inducible gene (designated ) responsible for loss of radiation resistance was identified. Investigations in vitro showed that the gene product of (PprA) preferentially bound to double-stranded DNA carrying strand breaks, inhibited exonuclease III activity, and stimulated the DNA end-joining reaction catalyzed by ATP-dependent and NAD-dependent DNA ligases. These results suggest that has a radiation-induced nonhomologous end-joining repair mechanism in which PprA plays a critical role.
