熱・流体・応力連成解析による水蒸気が及ぼす再冠水時のバリア性能への影響

本検討は、幌延深地層研究センターで実施中の人工バリア性能確認試験を対象に、熱・水・応力連成解析を実施した。再冠水時の熱・水・応力連成挙動は、間隙水の加熱により生じる水蒸気や、間隙圧の上昇により生じる気相の液相への溶解等の相変換の影響を受けると考えられる。そこで、相変換可能な熱・水・応力連成解析を実施し、水蒸気の発生や間隙空気の間隙水への溶解が再冠水挙動にどのような影響を及ぼすのか考察した。その結果、水蒸気を考慮することにより、ヒーター周辺の間隙率が増大し、乾燥密度の分布が生じた。さらに、気液二相流パラメータを変化させると緩衝材の応力状態が変化し、それに伴い、膨潤による緩衝材の膨出量にも影響を及ぼした。このため、緩衝材の長期挙動評価のうち再冠水挙動を模擬するには、相変換を考慮する必要があり、さらに、精緻な予測を行うためには、気液二相流パラメータの評価が重要であることが分かった。

Screening of cesium-accumulating mutant of radioresistant bacterium ${it Deinococcus radiodurans}$ by ion beam breeding technology

Bioremediation uses biological organisms to solve an environmental problem. However, microorganisms are sometimes not enough effective to achieve efficient recovery. Therefore, enhancing the functionality of microorganisms is needed to promote the active use of bioremediation. This study began with the purpose of development of Cs-accumulating bacteria by ion beam breeding technology. Previously, we suggested that ${it Deinococcus radiodurans}$ is the most suitable species for the development of high Cs-accumulating bacteria by ion beam breeding technology. Candidates of Cs-accumulating mutant were screened from the mutagenized cells by ion beam breeding technology on the basis of the luminescent intensity and the cell area as indicators of intracellular Cs level and cell growth, respectively. As a result, we obtained 33 candidates, whose Cs-accumulating abilities were over 2-fold higher than that of the wild-type, from the population of mutagenized cells.

アジア原子力協力フォーラム(FNCA)は、内閣府/文部科学省が主導する原子力平和利用協力の枠組みであり、その1つの活動として放射線育種プロジェクトがある。イネの突然変異育種の第二期プロジェクト(2013$$sim$$2017)として、バングラデシュ, 中国, インドネシア, 韓国, マレーシア, モンゴル, フィリピン, タイ, ベトナム及び日本の10ヶ国が参加して、「持続的農業のためのイネの品種改良(環境耐性、少肥料多収等)」が実施されている。バングラデシュでは、イオンビームを用いて高収量の新品種が数系統確立され、その内の1品種BINAdhan-14は2013年に品種登録され、実用栽培が開始されている。マレーシアでは、品種MR219への照射に由来する、渇水耐性や病害抵抗性の強い変異系統の選抜に成功し、実用化を予定している。ベトナムでは品種Khang danへの照射により、高収量で耐倒伏性の高い系統を獲得した。

A divertor simulation experimental module (D-module) with a V-shaped divertor target is installed in the west end-sell in GAMMA 10 large tandem mirror device, and a hydrogen plasma irradiation experiment to the target have been started to investigate radiation cooling mechanism on the target. A gas injection system is installed in the D-module and Langmuir probe and calorie meter array are mounted on the target plate. During the plasma irradiation, the highest electron density of 2.4 $$times$$ 10$$^{18}$$ m$$^{-3}$$ and a significant reduction of the electron temperature from a few tens of eV to 2 eV are achieved on the target plate by hydrogen and noble gas injection into the D-module.

イオンビーム育種技術は、近年植物や微生物の改変に活発に利用され、有用品種の作出に成果をあげている。東京電力福島第一原子力発電所の事故対応のためにイオンビーム育種でなにができるのか。日本原子力研究開発機構量子ビーム応用センターでは、高崎量子応用研究所のサイクロトロンを利用し、セシウム高・低吸収植物やセシウム高濃縮菌の開発に取り組み、変異体の候補を得ることに成功した。

Metabolome analysis of leaves of ${it Rumex obtusifolius}$ L. irradiated with ion beams

${it Rumex obtusifolius}$ (Polygonaceae) is a weed, which spreads all over the world. It contains amino acids and ascorbate (Vitamin C) more than other edible ${it Rumex}$ species (Miyagi et al., 2010). On the other hand, it highly accumulates soluble oxalate, which causes renal syndrome for vertebrates. Thus we aimed to lower oxalate contents in R. obtusifolius that can be used as a pasture by radiation breeding. In the present study, plant seeds irradiated with several doses of carbon ion beams ($$^{12}$$C$$^{6+}$$) were used to analyze effects of the radiation to oxalate contents and other metabolites using capillary electrophoresis-mass spectrometry (CE-MS). Results showed that the oxalate contents of leaves were increased by the irradiation of ion beams on the contrary to the case of the irradiation with $$gamma$$ rays. High oxalate plants tended to contain high levels of organic acids. Amino acids and phosphorylated organic acids were decreased in these plants.

Screening of salt tolerant mutants regenerated from irradiated stem segments of Lombardy poplar (${it Populus nigra}$)

Species of the genus ${it Populus}$ are ecologically important trees and often serving as a vegetative pioneer species because of their fast growth rates. The rapid growth of poplar is dependent on access to sufficient water in plantation sites and this is problematic. To overcome this problem, a forward genetics approach has been taken for developing salt/drought tolerant mutants by combination of radiation mutagenesis and in vitro regeneration in ${it Populus}$. Previously we screened regenerated ${it Populus nigra}$ shoots regenerated from stems exposed with cobalt-60 source of $$gamma$$ rays and isolated two mutant candidates named ${it g1}$ and ${it g5}$. Only about 25% stem segments of both lines produced roots in RIM with 75 mM NaCl. The low frequency in root production in NaCl medium may be attributed to that these plants are chimeric. We also successfully obtained at least 2 salt tolerant candidates regenerated from stem segments irradiated with 320 MeV $$^{12}$$C$$^{6+}$$ ion beams.

Development of new ${it Chrysanthemum Morifolium}$ pink mutants through ion beam irradiation

Developing new varieties of chrysanthemums is one of the emphases in Malaysian floriculture research as chrysanthemum is recognized as among the most important cut flower export for Malaysia. A combination of ion beam irradiation and ${it in vitro}$ organ culture techniques is very efficient in generating new ornamental varieties with novel traits and wider mutation spectrum. Therefore, in this project, ray floret cultures of pink chrysanthemum variety were irradiated at various doses with 320 MeV $$^{12}$$C$$^{6+}$$ ions, regenerated into plantlets and screened for morphological variations with an ultimate aim to produce mutants with useful horticultural traits. A series of screening was carried out at Nuclear Malaysia's glasshouse and MARDI Cameron Highlands. The mutants were selected and characterized morphologically. Based on these screenings, three mutants were identified based on their distinct flower and/or plant characteristics.

Alteration of segregation ratio suggests complex structural alteration of DNA induced by high-LET ion beams

大規模欠失を含む染色体の構造変化は、減数分裂過程での異常を引き起こすこと等により、後代への伝達を抑制すると考えられる。本研究では、突然変異の後代への伝達性のLET依存性に関する知見を得るため、葉の表面等の毛状突起の発達に必須である${it GL1}$遺伝子を指標として、照射した個体の後代における変異体の分離比を調査した。照射を行わなかった場合、材料であるシロイヌナズナの野生株と${it GL1}$遺伝子の変異株である${it gl1-1}$変異株を交配して得たF1種子の自殖後代では、野生型個体と無毛変異体は3対1の割合で出現すると期待される。平均LETが30keV/$$mu$$mのHeイオン、113keV/$$mu$$mのCイオン及び352keV/$$mu$$mのNeイオンを比較した場合、Neイオンでは系統毎の分離比の分布が期待値から大きく外れる個体が顕著に多く、高LETのイオンビームでは複雑な染色体の構造変化が起きていることが示唆された。

アジア原子力協力フォーラム(FNCA)は、内閣府/文部科学省が主導する原子力平和利用協力の枠組みであり、その活動の1つとして放射線育種プロジェクトがある。本プロジェクトでは、イネの突然変異育種を共通のテーマとして現在活動を展開している。2013年度から「持続的農業のためのイネの品種改良(環境耐性、少肥料多収等)」をテーマとし、10か国が参加して新たなプロジェクトが開始された。これまでの主要な成果として、バングラデシュでは、品種BRRIdhan29への炭素イオンの40Gy照射により、穂長が長く、多収の変異体を獲得した。インドネシアでは、炭素イオンの10Gyまたは20Gy照射により、早生で多収であるIB-3などの6系統の変異体を獲得した。マレーシアでは、品種MR219から、早生,穂数,千粒重,収量などに関する31の変異系統を獲得した。また、ベトナムでは、品種BacthomやKhandanへの照射により、収量や芳香に関する変異体の選抜を行っている。

This study began with the purpose of development of Cs-accumulating bacteria by ion beam breeding technology. We investigated which members are better suited for Cs-accumulation in the genus ${it Deinococcus}$ (${it D. radiodurans}$, ${it D. grandis}$, ${it D. proteolyticus}$, ${it D. radiopugnans}$, ${it D. geothermalis}$ and ${it D. murrayi}$) exhibit extraordinary radioresistant. The Cs concentration in the ${it Deinococcus}$ cells exhibited over 4-fold compared with that of the ${it E. coli}$ by atomic absorption spectrometer analysis. This result suggested that ${it D. radiodurans}$ and ${it D. grandis}$ are best two suitable species for the development of high Cs-accumulating bacteria by ion beam breeding technology.

Metabolic alterations in leaves of oxalate-rich plant ${it Rumex obtusifolius}$ L. irradiated by $$gamma$$ rays

We investigated on the metabolic effectiveness of $$gamma$$ ray irradiation to seeds of ${it Rumex obtusifolius}$, which are known to accumulate high level of soluble oxalate, to lower oxalate accumulation in leaves. Reduced rate of germination, discoloration of cotyledons, and deformed true leaves were observed by elevated irradiation. Metabolome analysis of primary metabolites using capillary electrophoresis-mass spectrometry showed a decrease in oxalate contents in the leaves of plants from $$gamma$$ ray-irradiated seeds. Moreover, organic acids such as malate and 2-oxoglutarate also decreased, whereas amino acids such as glutamate and glutamine increased. These results indicated that seed irradiation by $$gamma$$ rays leads to dynamic changes in metabolic pathways as well as plant growth/development.

Development of divertor simulation research in the GAMMA 10/PDX tandem mirror

In the large tandem mirror device GAMMA 10/PDX, a divertor simulation experimental module (D-module) with a V-shaped divertor target have been installed in the end-mirror. A massive gas injection of hydrogen and noble gases (argon and xenon) into the D-module during hydrogen plasma irradiation onto the target was performed, and plasma detachment from the target was investigated. Electron temperature measured by Langmuir probe array on the target was significantly reduced from a few tens of eV to $$<$$ 3 eV, and particle flux was also reduced. A bright H$$alpha$$ emission in the upstream region of the D-module and strong reduction near the target were observed by a two-dimensional image of H$$alpha$$ emission in the target observed with a high-speed camera. Molecular activated recombination (MAR) process is expected to contribute to the reduction of the electron temperature and the particle flux.

A forward genetics approach has been taken for developing salt/drought tolerant mutants by combination of radiation mutagenesis and ${it in vitro}$ regeneration in ${it Populus}$ since there is evidence that induced mutation with radiation mutagenesis has a great potential to produce new cultivars with wider mutation spectrum. Two mutant candidate shoots regenerated from $$gamma$$-ray irradiated stems were isolated by screening based on root induction ability on NaCl (75 mM)-supplemented rooting medium.

Generating new chrysanthemum plant varieties using ion beams

Chrysanthemum is among the most important cut flowers in Malaysia especially for export market. The main problem for chrysanthemum in Malaysia is the lack of new varieties to meet the ever-changing customer demands. To generate new chrysanthemum plant varieties using ion beams, this report discusses irradiation experiments for nodal cultures of chrysanthemum Pink variety. ${it In vitro}$ stems were cut into single nodes (0.5 cm in length) and cultured approximately 5 days before irradiation. Data on the survival of cultures, regenerated shoot height and number of leaves were taken 8 weeks after irradiation. The results suggested that the sensitivity of nodes to ion beam irradiation is similar to that of their ray floret counterparts examined last year.

Therefore, ${it R. obtusifolius}$ well growing in the meadow is a target of weeding. In this study, we aimed to develop a low oxalate ${it R. obtusifolius}$ plant that can be used as a pasture by radiation breeding. Plants from seeds irradiated with several doses of $$gamma$$ rays were used to analyze the radiation effect on oxalate content and other metabolite profiles. As a result, the oxalate content in leaves was decreased by $$gamma$$-ray irradiation. Plants with low oxalate content commonly contained increased level of amino acids and reduced level of organic acids.

Multidimensional thermal-hydraulic analysis on natural circulation behavior in ex-vessel fuel storage tank of MONJU

これまで「もんじゅ」の炉外燃料貯蔵槽(EVST)の過酷事故評価は1次元のフローネットワークコードSuper-COPDを用いて行われてきた。しかしEVST内の流体は自然循環によって駆動するため、過酷事故時の内部熱流動を正確に予想することは難しい。そこでEVST内部の熱流動挙動を明らかにし、フローネットワークモデルの適合性を評価するために多次元熱流動解析を実施した。この結果、EVST内の温度及び速度分布に関する多次元性の影響は小さく、上記のフローネットワークモデルの適合性はおおむね十分であることが示された。ただしフローネットワークモデルの回転ラック支持板の流動抵抗や冷却管の伝熱中心高さは、安全解析に対しては適切な範囲で保守的に設定する必要がある。

Generating new ornamental plant varieties using ion beams

Chrysanthemum is one of the leading cut flowers in Malaysia's cut flower industry. Due to the increasing demand of chrysanthemum cut flower, there is a need for new variety of chrysanthemum in order to compete to the ever-competitive cut flower market. Induced mutation breeding using ionizing radiation is one of the effective methods in plant breeding that resulted in generation of new mutated alleles and most unique feature. Attempt was carried out to apply mutation breeding technique using ion beam radiation to produce new variety of chrysanthemum. A pink variety of ${it Chrysanthemum morifolium}$ was used in this experiment. The experiment will be repeated to determine the accurate optimum irradiation dose for this variety. Meanwhile all shoots regenerated after irradiation are being sub-cultured and will be planted in the nursery.

エゾノギシギシへの$$gamma$$線照射効果

Therefore, ${it R. obtusifolius}$ well growing in the meadow is a target of weeding. In this study, we aim to develop a low oxalate ${it R. obtusifolius}$ plant that can be used as a pasture by radiation breeding. Plant seeds irradiated with several doses of $$gamma$$ rays were used to determine the germination rate and the lethal dose. As a result, the germination rate was not affected at doses up to 2000 Gy. However, the growth abnormality was shown in the lower dose. In the doses above 700 Gy, almost plants died after 1st leaf expansion. In conclusion, the 50% lethal dose (LD50) of $$gamma$$ rays was estimated to be 213 Gy for ${it R. obtusifolius}$.
