Concentration ratios of $$^{238}$$U and $$^{226}$$Ra for insects and amphibians living in the vicinity of the closed uranium mine at Ningyo-toge, Japan

There is still a scarcity of data on the transfer of naturally occurring radionuclides to wildlife in various ecosystems. In the present study, concentration ratios (CR$$_{rm wo-media}$$) of $$^{238}$$U and $$^{226}$$Ra were obtained for grasshoppers, frogs and newts in terrestrial and freshwater ecosystems. Soil, water and animal samples were collected for two years in the vicinity of the closed uranium mine at Ningyo-toge, Japan. Three sites with different $$^{238}$$U and $$^{226}$$Ra levels were of interest: (a) Pond and its shore (PO); (b) Low level stream and its shore near overburden dump (OD); (c) Uranium mill tailings pond and its shore (MP). The activity concentrations in both soil and water were PO $${approx}$$ OD $$<$$ MP for $$^{238}$$U, and PO $$<$$ OD $$<$$ MP for $$^{226}$$Ra. Regarding the wildlife, $$^{238}$$U was able to be determined for all samples, but the detection of $$^{226}$$Ra was observed only for a part of samples (not detected for grasshoppers at all). The means and standard deviations of CR$$_{rm wo-soil}$$ or CR$$_{rm wo-water}$$ were then calculated and may indicate the insignificant dependence of CR$$_{rm wo-media}$$ on environmental conditions characterized by the tested sites. The present data on CR$$_{rm wo-media}$$ were compared to the corresponding data or surrogate data from the IAEA's database, showing both agreement and discrepancy. Our data would contribute to enhancing the available data for those radionuclides and animals. In particular, the transfer to amphibians, which are one of main links in common food webs, is reported here for the first time.

Massive environmental monitoring has been conducted continuously after the Fukushima accident with different monitoring methods having different features together with migration studies of radiocesium in diverse environments. At three months after the accident, multiple radionuclides were detected at many places; while it was confirmed that radiocesium was most important from the viewpoint of long-term exposures. The air dose rates in environments related to human living have decreased faster than expected from radioactive decay by a factor of 2-3 on average. An empirical model for predicting air dose rate distribution was developed based on statistical analysis of massive car-borne survey data. Some trials were performed to integrate different types of contamination maps to obtain an integrated map of better quantity. Annual external exposure doses for residents who would return to their home were estimated to less than a few mSv as a whole. The environmental data and knowledge have been provided for diverse-spectrum of people in different ways.

重粒子線治療は、根治性の高さや患者への身体的負担の小さなガン治療法として成果を上げているが、患者には術後の2次発ガンのリスクが伴う。このリスク評価では、体内の放射線挙動や核反応の理解が不可欠となり、放射線輸送計算コードが有用なツールとなる。計算コードの重イオン核反応に対する検証は十分でないため、放射線生成過程の実験データが必要となる。そこで、本研究では、新しいガン治療用のビームとして使用されている核子あたり430MeVの炭素ビームと人体構成物質との核反応から放出される中性子の二重微分断面積の実験データを整備した。実験は、放射線医学総合研究所のHIMAC加速器において実施した。核子当たり430MeVの炭素ビームを45$$^{circ}$$に傾けた5cm$${times}$$5cm$${times}$$1cmの固体炭素標的に入射し、生成される中性子を15$$^{circ}$$, 30$$^{circ}$$, 45$$^{circ}$$, 60$$^{circ}$$, 75$$^{circ}$$および90$$^{circ}$$方向に設置した中性子検出器で測定した。また、中性子の運動エネルギーは飛行時間法により決定した。取得した実験データとPHITSの計算値を比較したところ、PHITSはこのエネルギー領域における炭素からの中性子生成を適切に模擬できることが分かった。

After the Fukushima Nuclear Power Plant accident in 2011 March, radiation doses have been measured for the residents by personal dosimeters and whole-body counters. In the current dose range, the potential radiation-induced risk could be small at the population level, however, should not be ignored. As suggested by NASA's reports on astronauts, the radiation sensitivity substantially varies depending on the individual genetic background. From the viewpoint of precision medicine, we note a possible issue: there might be the residents, including young children, with greater than average sensitivity because of their genetic background. Patients with DNA-damage-response defective disorders and the heterozygous carriers can be associated with the sensitivity as well as with cancer predisposition. We hereby propose that additional medical checkups for cancer, such as ultrasonography, gastrointestinal endoscopy, measurements of tumor markers in blood and urine, and genetic testing, should be combined in a balanced fashion to minimize their potential cancer risk in the future.

Reactions of edaravone (3-methyl-1-phenyl-2-pyrazolin-5-one) toward deoxyguanosine monophosphate (dGMP) hydroxyl radical adducts were investigated by pulse radiolysis technique. Edaravone was found to reduce the dGMP hydroxyl radical adducts through electron transfer reactions. The rate constants of the reactions were higher than 4 $$times$$ 10$$^{8}$$ dm$$^{3}$$ mol$$^{-1}$$ s$$^{-1}$$ and similar to those of the reactions of ascorbic acid, which is a representative antioxidant. Yields of single-strand breaks, base lesions, and abasic sites produced in pUC18 plasmid DNA by $$gamma$$ ray irradiation in the presence of low concentrations of edaravone were also quantified, and the chemical repair activity of edaravone was estimated by a method recently developed by the authors. By comparing suppression efficiencies to the induction of each DNA lesion, it was found that base lesions and abasic sites were suppressed by the chemical repair activity of edaravone, although the suppression of single-strand breaks was not very effective. This phenomenon is attributed to the chemical repair activity of edaravone toward base lesions and abasic sites. However, the chemical repair activity of edaravone for base lesions was lower than that of ascorbic acid.

Order effect of repair processes to clustered DNA damage

In a living cell, cluster DNA damage is thought to be processed by several different pathways simultaneously or sequentially. Under this situation the cellular response to cluster DNA might depend on the order of repair processes because the configuration of the lesions will be modified by the reaction of initial repair protein, affecting the DNA-binding or excision activities of latter proteins. In the present study, we investigate whether initial enzymatic repair affects latter processes. Plasmid DNA exposed to C ion is treated with two base excision repair enzymes, Nth and Fpg, which convert pyrimidine and purine lesions to a SSB, respectively. Obtained results show that the amount of enzymatically induced SSB is very slightly less in DNA sample treated with Nth first and then Fpg than that of other treatments. These results indicate that the configuration-change of the cluster by the first enzymatic treatment does not significantly influence the activity of secondary enzyme.


Dependence of the yields of AP sites and AP clusters produced in plasmid DNA on scavenging capacity and LET

The object of the study is to reveal whether the induction process of an abasic site (AP site) and the clustered DNA damage which contain AP sites (AP cluster) depends on ionizing density of radiation. In order to clarify the relation between track structure of C ions and the induction processes of an AP site or AP cluster, we measure the yield of AP using Nfo protein as an enzymatic probe, which converts an AP site to detectable single strand break. Several scavenging capacities in the samples are tested to estimate the effect of OH radicals. We compare those experimental yields with theoretical ones that are obtained from Monte Carlo track simulation to develop a model of AP site induction by irradiation. The yields of AP sites analyzed by the simulation are in good agreement with experimental ones. As a result, we newly determined the branching ratio of induction of the AP the site in the simulation.


Live-cell imaging study of mitochondrial morphology in mammalian cells irradiated

In this study, we report a new approach of tracing the mitochondrial morphological change of the cells exposed to high LET radiation using a live-cell imaging technique. We used the NMuMG (Normal murine mammary gland)-FUCCI2 cell line. Using the FUCCI2 expressing cells, we can observe red fluorescence in the nuclei of G1 phase cells and green fluorescence in the nuclei of S/G2/M phase cells. We labeled mitochondria by Mitotracker Red. Kinetics of mitochondrial morphology was analyzed by the live-cell imaging technique using a fluorescence microscope. Mitochondrial images were captured for 4 days after irradiation. In a preliminary study, we found that X-ray irradiation of cells caused mitochondrial fragmentation, and proportion of cell population with fragmented mitochondria increased with increasing dose and with time after irradiation. It is demonstrated that the method proposed in this study works successively to trace cytoplasmic effects by high LET irradiation.


Visualization of cell-cycle modification by ionizing irradiation in single HeLa cells using fluorescent ubiquitination-based cell-cycle indicator

Fluorescent ubiquitination-based cell cycle indicator (FUCCI) HeLa cells are one of useful model cell lines to visualize a cell-cycle because their nuclei show different colors; orange indicating G1; green indicating S/G2. In order to establish a novel assay system to study cell-cycle modification by high LET irradiation such as ion beams for cancer therapy we have observed time-lapse images of HeLa-FUCCI cells irradiated as a preliminary experiment using conventional X-rays instead of high LET ion beams. The cell-cycle was strongly arrested by irradiation at S/G2 and never progressed to G1. In contrast, cells irradiated at G1 progress to S/G2 with a similar time course as non-irradiated control cells. These results show that single FUCCI cell exposure and live cell imaging are a powerful method to trace the single cell effect of high LET irradiation on the cell-cycle in future.
